Abstract

INDONESIA:

Diabetes melitus (DM) memiliki komplikasi berupa kardiomiopati diabetikum yang dipicu oleh stres oksidatif yang dapat meningkatkan kadar laktat dehidrogenase (LDH) pada jantung. Ekstrak daun Clinacanthus nutans berpotensi sebagai antioksidan karena memiliki kandungan fitokimia. Penelitian ini bertujuan untuk mengetahui adanya pengaruh pemberian ekstrak daun Clinacanthus nutans terhadap kadar LDH jantung pada tikus yang diinduksi streptozotocin (STZ). Sebanyak 30 ekor tikus dibagi menjadi 5 kelompok. Kelompok kontrol normal (K1) tidak diberi perlakuan. Kelompok kontrol DM (K2) diinjeksi STZ dan tidak diberi perlakuan. Kelompok perlakuan 1, 2, dan 3 adalah kelompok perlakuan diinjeksi STZ dengan penyondean berturut-turut ekstrak daun Clinacanthus nutans dosis 100, 200, dan 400 mg/kgBB. Pada khir penelitian, tikus diterminasi dan sampel darah diambil dengan cara pungsi dari jantung dan diuji kadar LDH menggunakan spektrofotometer UV-Vis dengan panjang gelombang 450 nm. Analisis data menggunakan uji non-parametrik Kruskal-Wallis, dan dilanjutkan dengan uji Post Hoc Mann-Whitney. Hasil penelitian menunjukkan ekstrak daun Clinacanthus nutans mampu menurunkan kadar LDH jantung tikus model DM secara signifikan dibandingkan dengan tanpa pemberian perlakuan. Kesimpulan dari penelitian ini, pemberian ekstrak etanol 70% daun Clinacanthus nutans berpengaruh dalam menurunkan kadar LDH jantung tikus model DM.

Diabetes mellitus (DM) has complications in the form of diabetic cardiomyopathy triggered by oxidative stress which can increase lactate dehydrogenase (LDH) levels in the cardiac. Clinacanthus nutans leaf extract has the potential as an antioxidant because it contains phytochemical compounds. This study aims to determine the effect of Clinacanthus nutans leaf extract on cardiac LDH levels in streptozotocin (STZ) induced rats. A total of 30 rats were divided into 5 groups. The normal control group (K1) was not given any treatment. The DM control group (K2) was injected with STZ and not given any treatment. Treatment groups 1, 2, and 3 were injected with STZ and then administered Clinacanthus nutans leaf extract at doses of 100, 200, and 400 mg/kgBW, respectively. At the end of the study, the rats were terminated and blood samples were collected by cardiac puncture and tested for LDH levels using a UV-Vis spectrophotometer at a wavelength of 450 nm. Data analysis used the non-parametric Kruskal Wallis test, followed by the Post Hoc Mann-Whitney test. The results showed that Clinacanthus nutans leaf extract was able to significantly reduce cardiac LDH levels in DM rat models compared to those without treatment. The conclusion of this study is that administration of 70% ethanol extract of Clinacanthus nutans leaves has an effect in lowering cardiac LDH levels in DM rat models.
