Abstract

INDONESIA:

Diabetes melitus ditandai oleh hiperglikemia kronis yang dapat memicu stres oksidatif pada berbagai organ, termasuk testis. Stres oksidatif meningkatkan peroksidasi lipid dan menghasilkan malondialdehid (MDA) sebagai produk akhir yang berperan sebagai penanda kerusakan sel. Clinacanthus nutans mengandung senyawa fitokimia yang memiliki aktivitas antioksidan potensial. Penelitian ini bertujuan untuk mengevaluasi pengaruh ekstrak etanol daun Clinacanthus nutans terhadap kadar MDA testis tikus Wistar model diabetes melitus yang diinduksi streptozotosin (STZ). Sebanyak 30 ekor tikus jantan dibagi secara acak menjadi lima kelompok, yaitu kontrol normal (K1), kontrol diabetes (K2), serta tiga kelompok perlakuan (P1, P2, P3) yang menerima ekstrak etanol daun C. nutans dengan dosis 100, 200, dan 400 mg/kgBB setelah induksi STZ. Ekstrak diberikan secara oral selama 28 hari berturut-turut. Setelah perlakuan, tikus diterminasi, dan jaringan testis diambil untuk analisis kadar MDA menggunakan metode Thiobarbituric Acid Reactive Substances (TBARS) dengan spektrofotometer UV-Vis pada panjang gelombang 532 nm. Data dianalisis menggunakan uji Kruskal Wallis yang dilanjutkan dengan uji Mann Whitney. Hasil penelitian menunjukkan bahwa pemberian ekstrak etanol daun C. nutans secara signifikan menurunkan kadar MDA testis dibandingkan kelompok kontrol diabetes. Temuan ini menunjukkan bahwa ekstrak etanol daun Clinacanthus nutans memiliki efek antioksidan melalui penurunan peroksidasi lipid dan stres oksidatif pada testis tikus model diabetes melitus.

ENGLISH:

Diabetes mellitus is characterized by chronic hyperglycemia that induces oxidative stress in multiple organs, including the testis. Oxidative stress enhances lipid peroxidation and produces malondialdehyde (MDA) as a final product, which serves as a biomarker of cellular damage. Clinacanthus nutans leaves are known to contain phytochemical compounds with antioxidant activity. This study aimed to evaluate the effect of the 70% ethanolic extract of C. nutans leaves on testicular MDA levels in streptozotocin-induced diabetic Wistar rats. Thirty male rats were randomly assigned into five groups: normal control (K1), diabetic control (K2), and three treatment groups (P1, P2, P3) that received C. nutans extract at doses of 100, 200, and 400 mg/kg body weight, respectively, following STZ induction. The treatment was administered orally for 28 consecutive days. After treatment, the rats were sacrificed, and testicular tissues were collected for MDA analysis using the Thiobarbituric Acid Reactive Substances (TBARS) method with a UV-Vis spectrophotometer at 532 nm. Data were analyzed using the Kruskal Wallis test followed by the Mann–Whitney post hoc test. The results demonstrated that administration of C. nutans extract significantly decreased testicular MDA levels compared with the diabetic control group. These findings suggest that the 70% ethanolic extract of Clinacanthus nutans leaves exerts antioxidant effects by attenuating oxidative stress in the testis of diabetic rat models.
