Abstract
INDONESIA:
Pendahuluan: Influenza merupakan penyakit infeksi saluran pernapasan yang masih menjadi masalah kesehatan global akibat tingginya angka morbiditas dan mortalitas serta meningkatnya kasus resistensi terhadap antivirus konvensional seperti oseltamivir. Oseltamivir bekerja dengan menghambat aktivitas enzim neuraminidase yang digunakan oleh virus influenza untuk keluar dari sel yang terinfeksi. Biji kopi Arabika diketahui mengandung senyawa bioaktif yang memiliki aktivitas farmakologis, termasuk potensi sebagai agen antivirus. Tujuan: Mengevaluasi potensi senyawa dalam biji kopi Arabika sebagai agen anti-influenza secara in silico dengan menargetkan enzim neuraminidase virus influenza. Metode: Lima senyawa aktif dalam kopi Arabika (asam kafeat, asam klorogenat, asam kinat, asam malat, dan asam sitrat) dianalisis menggunakan molecular docking terhadap neuraminidase (PDB ID: 5NWE). Evaluasi farmakokinetik dilakukan menggunakan SwissADME berdasarkan hukum Lipinski dan prediksi absorpsi usus (HIA), serta uji toksisitas dilakukan dengan preADMET, dan prediksi aktivitas biologis dengan PASS. Hasil: Asam klorogenat dan asam kafeat menunjukkan nilai afinitas pengikatan yang kuat terhadap neuraminidase, masing-masing sebesar −8 dan −6,3 kkal/mol, mendekati afinitas oseltamivir yaitu −6,3 kkal/mol. Sesuai dengan hasil prediksi aktivitas biologis dengan hasil asam klorogenat dan asam kafeat masing-masing sebesar 0.549 dan 0.537 yang termasuk ke dalam kategori sedang. Kesimpulan: Senyawa dalam biji kopi Arabika berpotensi sebagai agen anti-influenza alami yang efektif dan aman berdasarkan prediksi fisikokimia, farmakokinetik, farmakodimanik, dan hasil molecular docking yang telah dilakukan.

Introduction: Influenza is a respiratory infectious disease that remains a global health concern due to its high morbidity and mortality rates, as well as the increasing resistance to conventional antivirals such as oseltamivir. Oseltamivir works by inhibiting the activity of the neuraminidase enzyme used by influenza viruses to exit infected cells. Arabica coffee beans are known to contain bioactive compounds with pharmacological activities, including potential antiviral properties. Objective: To evaluate the potential of compounds in Arabica coffee beans as anti-influenza agents through an in silico approach by targeting the influenza virus neuraminidase enzyme. Methods: Five active compounds found in Arabica coffee (caffeic acid, chlorogenic acid, quinic acid, malic acid, and citric acid) were analyzed using molecular docking against neuraminidase (PDB ID: 5NWE). Pharmacokinetic evaluation was conducted using SwissADME based on Lipinski's Rule of Five and intestinal absorption (HIA) prediction, toxicity tests were performed using preADMET, and biological activity prediction was conducted using PASS. Results: Chlorogenic acid and caffeic acid showed strong binding affinity to neuraminidase, with values of −8 and −6.3 kcal/mol, respectively, close to the affinity of oseltamivir (−6.3 kcal/mol). Based on biological activity prediction, chlorogenic acid and caffeic acid obtained values of 0.549 and 0.537, respectively, categorized as moderate activity. Conclusion: Compounds in Arabica coffee beans show potential as safe and effective natural anti-influenza agents based on physicochemical, pharmacokinetic, pharmacodynamic predictions, and molecular docking results.
