Abstract
INDONESIA:
Kanker otak merupakan salah satu penyebab kematian. Sel penyusun otak yang berpotensi untuk mengalami kanker adalah sel neuroglia. Salah satu senyawa karsinogen penyebab kanker adalah 7,12 dimetilbenz (α) antrasen (DMBA). Beberapa penelitian telah dilakukan untuk pengobatan penyakit kanker dengan cara mengisolasi senyawa antikanker dari bahan alam seperti tumbuh-tumbuhan. Salah satu tumbuhan yang berpotensi sebagai obat kanker adalah Sirsak (Annona muricata L.). Sirsak memiliki senyawa aktif yang berpotensi sebagai antikanker yaitu Annonaceous Acetogenin. Penelitian ini bertujuan untuk mengetahui pengaruh dan nilai LC50 pemberian ekstrak etanol daun sirsak (Annona muricata L.) terhadap pertumbuhan sel neuroglia baby hamster yang dipapar dengan 7,12-dimetilbenz (α) antrasen (DMBA) dalam kondisi in vitro.
Penelitian ini menggunakan sel neuroglia yang diambil dari baby hamster umur dua hari yang ditumbuhkan dalam medium dasar DMEM (Dulbecco’s Modified Eagle’s Medium) dengan dan tanpa dimetilbenz (α) antrasen (DMBA). Konsentrasi ekstrak etanol daun sirsak yang digunakan adalah 0 µg/ml, 10 µg/ml, 20 µg/ml, 40 µg/ml, 80 µg/ml dan 160 µg/ml. Parameter dalam penelitian ini adalah konfluen, viabilitas dan uji sitotoksisitas dengan menggunakan analisis probit sehingga didapatkan LC50.
Hasil penelitian menunjukkan bahwa, pemberian ekstrak etanol daun sirsak (Annona muricata L.) berpengaruh terhadap pertumbuhan sel neuroglia baby hamster yang dipapar dengan 7,12-dimetilbenz (α) antrasen (DMBA) dalam kondisi in vitro. Pemberian ekstrak daun sirsak mampu menurunkan persentase konfluen, meningkatkan persentase kematian dan bersifat sangat toksik terhadap sel kanker berdasarkan hasil analisis probit dengan nilai LC50 sebesar 27,64 µg/mL

Brain cancer is one of the causes of death. Tues brain cell constituent that has the potential to develop cancer are neuroglia cells. One of the compounds carcinogen was 7.12 dimetilbenz (α) antrasen (DMBA). Several studies have been conducted for the treatment of cancer by means of anticancer compounds isolated from natural ingredients such as herbs. One of the plants that have the potential as a cancer drug is Soursop (Annona muricata L.). Soursop has a potentially active compounds as anticancer is Annonaceous acetogenin. This study aimed to investigate the effect and LC50 values of soursop (Annona muricata L.) leaf ethanol extract on the growth of neuroglia cells baby hamster were exposed to 7.12-dimetilbenz (α) antrasen (DMBA) in vitro conditions.
This study uses neuroglia cells taken from baby hamster two days old grown in basic medium DMEM (Dulbecco's Modified Eagle's Medium) with and without dimetilbenz (α) antrasen (DMBA). Concentration of Soursop leaf ethanol extract used is 10 µg/ml, 20 µg/ml, 40 µg/ml, 80 µg/ml and 160 µg/ml. The parameters in this study were confluent, viability and cytotoxicity assay using probit analysis to obtain the LC50.
The results showed that the ethanol extract of soursop leaf (Annona muricata L.) having an effect on the growth of neuroglia cells baby hamster were exposed with 7.12- dimetilbenz (α) antrasen (DMBA) in vitro conditions. Soursop leaf ethanol extract can lower percentage of confluent, increase the percentage of deaths and is highly toxic to the cancer cells by probit analysis with LC50 values of 27.64 µg/mL.
