Abstract
ABSTRAK
Kambing Saanen (Capra aegagrus H.) merupakan kambing tipe perah yang mempunyai nilai nutrisi yang hampir sama dengan susu sapi, bahkan diyakini bahwa susu kambing mempunyai nilai lebih dibandingkan dengan susu sapi. Susu kambing terkenal karena kandungan nilai nutrisi dan nilai medinya sejak zaman dahulu. Kambing Saanen ini mempunyai rata rata produksi susu tertinggi dibandingkan bangsa kambing manapun.Rata-rata produksi susu kambing Saanen di daerah tropis adalah 1 – 3 kg per hari, sedangkan di daerah subtropis dapat mencapai 5 kg per hari. Kambing PE adalah kambing perah, yaitu hasil persilangan antara kambing kacang betina dengan kambing Etawah jantan. Kambing PE digolongkan sebagai kambing tipe dwiguna yaitu sebagai penghasil daging dan susu. Produksi susu kambing PE berkisar antara 0.5-0,7 liter/ekor/hari. Bakteri asam laktat merupakan jenis bakteri yang mampu menghasilkan asam laktat, hidrogen peroksida, antimikroba dan hasil metabolisme lain yang memberikan pengaruh positif bagi produktivitas. Tujuan penelitian ini adalah untuk mendapatkan informasi keragaman dan karakteristik bakteri asam laktat yang terdapat pada susu kambing Saanen (Capra aegagrus H.)
Penelitian ini dilakukan secara deskriptif untuk mengetahui adanya bakteri asam laktat (BAL) dari susu kambing Saanen (Capra aegagrus H.). proses fermentasi dilakukan pada media MRS broth dengan suhu 37º C selama 24 jam, sampel kemudian diencerkan bertingkat yang bertujuan untuk mengurangi kepadatan koloni yang tumbuh pada media isolasi. Pengenceran dilakukan pada media MRS agar dengan media tuang (pour plate) yang diinkubasi pada suhu 37º C selama 48 jam. Tiap koloni yang berbeda dimurnikan dengan metode gores. Didapatkan 3 isolat yaitu SP1 SP2 SP3. Kemudian dilakukan karakterisasi BAL dengan pewarnaan gram dan pewarnaan endospora serta uji katalase. Kemudian diidentifikasi menggunakan Microbact 12B dan didapatkan 1 isolat yang teridentifikasi Khamir yaitu SP 1 dan 2 isolat teridentifikasi BAL yaitu SP2 dan SP3.
Hasil penelitian menunjukkan bahwa dari susu kambing Saanen (Capra aegagrus H.) didapatkan 1 isolat yang bersifat Gram negatif, endospora negatif, katalase negatif, serta uji identifikasi dengan microbact merupakan termasuk kategori khamir. Selanjutnya 2 isolat bakteri yang bersifat Gram positif, endospora negatif, katalase negatif, serta uji identifikasi dengan microbact merupakan termasuk kategori bakteri asam laktat yaitu Lactobacillus bulgaricus dan Lactobacillus desidiosus.
ABSTRACT
Saanen Goat ( Capra aegagrus H.) is the type of dairy goats which has value nutrition similar to cow milk ,but it was believed that goats milk more valueable than cow milk .Goats milk has been famous in the past because their nutrition and medical values. Saanen Goat has the higest milk production compared with the others goats .The average of Saanen Goat milk production about 1-3 pounds per day in the tropics, while in the subtropics can reach 5 pounds per day . PE goat included of type dairy goats , which namely as the results of a cross between the nut female goats with etawah male goats .PE goats has been classification into the two functions type products, the meat and dairy productions .PE goats milk production reach about 0.5-0,7 liter / tail / day .Lactic acid bacteria is the type of bacteria which capable to producing lactic acid, hydrogen peroxide, antimicroba, some results metabolism which give a positive influence for dairy productivity. The purpose of this research is to get information about diversity and characteristic of lactic acid bacteria which is found in the saanen goat milk (Capra aegagrus H.)
The purpose of this research to knowing description about lactic acid bacteria (LAB) from saanen goat milk (capra aegagrus H.). The process of this research begin from fermentation stage which performed on the MRS broth media with boiling products in temperature 37º celcius degree for 24 hours, then the sample diluted in graded, which aimed to minimize the density of a growing colony in isolation media. the Dilution processed on MRS jelly media (pour plate) which incubate at the 37º celcius degree for 48 hours. Each different colonies purified by plate methode. Then, it gain 3 isolates namely sp1 sp2 sp3. the process continued with characterization step of lactac acid bacteria using staining grams and staining endospores and catalase test. Then, identified using microbact 12b and obtained 1 identified isolates namely Khamir as sp 1 and 2, which isolates identified lactac acid bacteria namely sp2 and sp3.
The result of this research showed 2 important conclutions, First, saanen goats milk (Capra aegagrus H .) classified into Khamir category which obtained from 1 isolates using microbacteria method, has characteristics like Gram negative, negative endospores, and negative catalase. Second, saanen goats milk also identified 2 isolates included lactic acid bacteria (Lactobacillus bulgaricus and Lactobacillus desidiosus) which obtained from microbact method which has characteristics different from the first. They are identified as gram positive , negative endospores, and negative catalase.
