Abstract
ABSTRAK
Bekatul merupakan salah satu limbah pertanian yang mengandung selulosa 40-60%. Kandungan selulosa yang cukup tinggi pada bekatul dapat dimanfaatkan sebagai substrat dalam pembuatan glukosa melalui hidrolisis selulosanya. Hidrolisis enzimatis menggunakan ekstrak kasar enzim selulase menjadi salah satu alternatif sebagai upaya mengurangi jumlah limbah pertanian dan memenuhi kebutuhan glukosa yang makin meningkat tiap tahunnya.
Tahapan awal bekatul direndam dalam larutan NaOH 1 % selama 2 jam dan dilakukan tahapan hidrolisis menggunakan ekstrak kasar enzim selulase. Tahapan hidrolisis menggunakan metode Rancangan Acak Lengkap (RAL) yang terdiri dari dua faktor yaitu konsentrasi substrat 2; 4; 6; 8 dan 10 % (b/v) dan lama inkubasi 12; 18 dan 24 jam. Data hasil hidrolisis dianalisis menggunakan analisis varians (ANOVA) dan uji BNT (Beda Nyata Terkecil).
Kadar glukosa awal setelah perendaman dengan NaOH 1% sebesar 12,784 ppm. Kondisi optimum hidrolisis yaitu pada konsentrasi substrat 8 % dan inkubasi 24 jam dengan perolehan yield glukosa sebesar 0,152 % dan kadar glukosa sebesar 121,836 ppm. Hasil analisis menggunakan uji ANOVA (α=1 %) menunjukkan bahwa konsentrasi substrat dan lama inkubasi berpengaruh nyata pada kadar glukosa. Uji BNT menyatakan perlakuan C4T3 dan C5T3 dengan hasil glukosa 121,836 ppm dan 142,673 ppm merupakan perlakuan yang berbeda nyata.
ABSTRACT
Rice bran is kind of agriculture waste which content of cellulose 40-60 %. The cellulose of rice bran can be used for substrate at production procces of glucose by hydrolysing its cellulose. Hydrolysis enzymatic use the cellulase crude enzyme is one of the effort to reduce agriculture waste and complete the glucose requirement that increase every years.
The pretreatment process of rice bran was dissolved into NaOH 1 % solution for 2 hours and the hydrolysis step used crude enzyme of cellulase. The hydrolysis process was using the Complete Random Design (CRD) method which consist of two factors were substrate concentration i.e 2; 4; 6; 8 and 10 % (w/v) and incubation time i.e 12; 18 and 24 hours. The data of hydrolysis result was analysed using varians analysis (ANOVA) and LSD (Least Significant Difference) % test.
The concentration of glucose was 12,784 ppm. The optimum condition of hydrolysis was obtained on substrate concentration 8 % and incubation time 24 hours which yield of glucose 0,152 % and concentration of glucose 121,836 ppm. The result of ANOVA (α=1 %) test showed the treatment of hydrolysis process was given a significant different to concentration of glucose. The LSD test showed the treatments C4T3 and C5T3 with result of glucose i.e 121,836 ppm and 142,673 ppm were the significant different of treatments.
