Abstract
ABSTRAK
Malaria adalah penyakit infeksi parasit menular yang disebabkan oleh Plasmodium, yang tersebar luas dan paling dikenal karena banyak menimbulkan kematian sepanjang zaman. Telah dilakukan penelitian tentang uji efektivitas antimalaria dan identifikasi golongan senyawa aktif ekstrak etanol 80% tanaman Anting-anting (Acalypha indica L.) pada mencit yang terinfeksi plasmodium berghei. Penelitian ini bertujuan untuk memanfaatkan bahan alam sebagai antimalaria alami, dengan menentukan efektivitasnya dalam menghambat parasit P.berghei dan mengetahui kandungan senyawa aktif menggunakan KLT. Allah telah menjelaskan dalam al Quran surat asy Syu’ara ayat 7 bahwa Maha Besar Allah yang mana telah menciptakan berbagai macam tumbuhan yang baik dan bermanfaat. Penelitian ini ingin mengetahui bahwa tanaman Anting-anting berpotensi memberikan manfaat sebagai antimalaria.
Uji efektivitas antimalaria dilakukan secara in vivo. Hewan coba mencit diinfeksi dengan 106 parasit Plasmodium berghei dan dibagi dalam 8 kelompok perlakuan, (1) Kontrol non infeksi; (2) Kontrol negatif; (3) Kontrol positif (klorokuin dosis 5,71 mg/kg BB); (4) Anting-anting dosis 0,01 mg/kg BB; (5) Anting-anting dosis 0,1 mg/kg BB; (6) Anting-anting dosis 1 mg/kg BB; (7) Anting-anting dosis 10 mg/kg BB; dan (8) Anting-anting dosis 100 mg/kg BB. Perlakuan dimulai pada hari ke-0 ketika derajat parasitemia mencapai 1 – 5% dan sampai 4 hari terapi. Kemudian dilanjutkan uji fitokimia menggunakan reagen dan diperkuat dengan identifikasi golongan senyawa aktif menggunakan Kromatografi Lapis Tipis.
Hasil uji efektivitas antimalaria secara in vivo menunjukkan nilai % penghambatan mulai dosis 0,01 mg/kg BB; 0,1 mg/kg BB; 1 mg/kg BB; 10 mg/kg BB; dan 100 mg/kg BB secara berurutan adalah 25,50%; 29,20%; 31,30%; 58,11%; dan 65,22%. Berdasarkan % penghambatan tersebut diperoleh nilai ED50 sebesar 6,32 mg/kg BB dengan nilai Pearson correlation sebesar 0,775. Kemudian kandungan senyawa aktif yang diperoleh adalah golongan terpenoid, alkaloid dan flavonoid.
ABSTRACT
Malaria is an infective disease parasite caused by Plasmodium spread widely and mostly well-known since they are a big cause of death from time to time. Many researches have been conducted to test the effectiveness of anti-malaria and identification of active compound classification of 80% ethanol extract contained in Acalypha indica L in Mouse infected with Plasmodium berghei. This research was aimed to utilize nature material to be used as natural anti-malaria by determining its effectiveness impede P. Berghei parasite and find out the content of active compound by using KLT. Allah has explained in Quran, surah Asy syu’ara verse 7 that the Almighty God creating any kinds of good and advantageous plants. This research was intently conducted to find out if Acalypha indica L had potential to be anti-malaria.
Effectiveness test of anti-malaria was conducted in an in vivo method. An experimental mouse was infected with 106 of Plasmodium berghei parasite and divided into 8 different treatment (1) non-infected control; (2) negative control; (3) positive control (5.71 mg/kg BB of kloroku in-dose); (4) 0.01 mg/kg BB of Acalypha indica L’s dose; (5) 0.1 mg/kg BB of Acalypha indica L’s dose; (6) 1 mg/kg BB of AcalyphaindicaL’s dose; (7) 10 mg/kg BB of Acalypha indica L’s dose; and (8) 100 mg/kg BB of Acalypha indica L’s dose. The treatments were started in on day 0 when parasitemia parasite degree reached 1-5% and until 4 days of therapy. Then, a phytochemical test was conducted by using reagent and it was strengthened by identifying classification of active compound by using thin layer Chromatography.
The result of anti-malaria effectiveness conducted in in vivo method showed that the % value of impediment for each dose of 0.01 mg/kg BB; 0.1 mg/kg BB; 1 mg/kg BB; 10 mg/kg BB; dan 100 mg/kg BB were respectively: 25,50%; 29,20%; 31,30%; 58,11%; dan 65,22%. In accordance with % of the impediment, a value amount of 6.32 mg/kb BB with Pearson correlation value of 0.775 was resulted. In addition, the content of active compound resulted was terpenoid, alkaloid, and flavonoid group.
