Abstract
ABSTRACT
Phenylketonuria is a genetic disease caused by deficiencies or inexistence hidroxylase phenylalanine enzyme (PAH) which has function in changing phenylalanine amino acid into tyrosine. The high concentration of phenylalanine in body will be converted into phenylpyruvic acid and released through urine. The purpose of this research is looking for method to analyze phenylketonuria using silica gel plate immobilized with FeCl3 reagent, MgSO4 reagent and CH3COONa-HCl buffer.
This research consists of sample preparation, materials preparation, determination the best concentration of FeCl3 and MgSO4 reagents, and optimum pH of CH3COONa-HCl buffer, immobilizes FeCl3, MgSO4, and CH3COONa-HCl buffer into silica gel plate to identify phenylpyruvate, determination of smallest concentration of phenylpyruvate in water and healthy baby urine solvents with immobilized silica gel plate by phenylpyruvate identified reagent method and data analyze.
The result of this research shows the best concentration FeCl3 and MgSO4 reagents is 3 mmol/L FeCl3 and 3 mmol/L MgSO4, while optimum pH of CH3COONa- HCl buffer is 2,0. The best immobilization technique is soaking silica gel plate into identified phenylpyruvate reagent in 30 minutes and drying it in oven at temperature 35 oC in 10 minutes. The smallest concentration of phenylpyruvate in water and healthy baby urine solvents which able to detected show the same result that is sodium phenylpyruvate 0,5 mmol/L.
ABSTRAK
Phenylketonuria ialah suatu penyakit keturunan yang disebabkan karena defisiensi atau tidak adanya enzim fenilalanin hidroksilase (PAH) yang merubah asam amino fenilalanin menjadi tirosin. Kadar fenilalanin yang tinggi pada tubuh akan dikonversi menjadi asam fenilpiruvat dan dikeluarkan lewat urine. Tujuan penelitian ini mencari metode analisis phenylketonuria menggunakan plat silika gel yang terimmobilisasi reagen FeCl3, MgSO4 dan larutan buffer CH3COONa-HCl.
Penelitian ini meliputi preparasi sampel, preparasi bahan, penentuan jumlah mol terbaik reagen FeCl3, MgSO4 dan pH optimum larutan buffer CH3COONa-HCl, immobilisasi terbaik reagen FeCl3, MgSO4 dan larutan buffer CH3COONa-HCl pada plat silika gel untuk identifikasi fenilpiruvat, penentuan konsentrasi terkecil fenilpiruvat pada pelarut air dan urine bayi sehat dengan metode plat silika gel terimmobilisasi reagen identifikasi fenilpiruvat dan analisis data.
Hasil penelitian menunjukkan jumlah mol terbaik reagen FeCl3 dan MgSO4 untuk bereaksi dengan fenilpiruvat secara spektrofotometri tampak adalah FeCl3 3 mmol/L dan MgSO4 3 mmol/L, sedangkan pH optimum larutan buffer CH3COONa-HCl adalah pH 2,0. Teknik immobilisasi reagen FeCl3, MgSO4 pada mol terbaik dan larutan buffer pada pH optimum diperoleh pada waktu perendaman plat silika gel selama 30 menit dan pengeringan pada suhu 35 oC selama 10 menit. Konsentrasi terkecil fenilpiruvat pada pelarut air dan urine bayi sehat yang dapat terdeteksi dengan metode plat silika gel terimmobilisasi reagen identifikasi fenilpiruvat menunjukkan hasil yang sama yaitu natrium fenilpiruvat 0,5 mmol/L.
