Abstract
ABSTRAK
Pb asetat merupakan salah satu sumber dari radikal bebas yang dapat meningkatkan ROS sehingga menjadi tidak terkontrol dan menyebabkan penurunan infertilitas pada pria. Karena menyebabkan stress oksidatif yang akan menyebabkan berkurangnya motilitas, viabilitas dan kerusakan akrosom spermatozoa sehingga dapat merusak enzim superoksida-dismutase (SOD). SOD bertugas dalam mempertahankan fungsi sel sehingga fungsi sel menurun dan menjadi rusak. Oleh karena itu tujuan penelitian ini untuk mengetahui pengaruh pemberian minyak jintan hitam terhadap kualitas spermatozoa dan kadar superoxide dismutase SOD testis mencit yang dipapar Pb asetat peroral.
Penelitian ini merupakan penelitian eksperimental yang menggunakan Rancangan Acak Lengkap (RAL) dengan 4 kali perlakuan dan 5 kali ulangan. Hewan coba yang digunakan adalah mencit strain balb/c jantan berumur 2-3 bulan yang berjumlah 20 ekor. Perlakuan dalam penelitian ini meliputi pemaparan timbal (Pb) 0,3 mg/grBB/hari/oral selama 5 hari, pemberian minyak jintan hitam dengan dosis 0,6 mg/grBB, dan dosis 1,2 mg/grBB selama 10 hari. Hasil yang diamati dalam penelitian ini adalah kualitas spermatozoa dan kadar superoxide dismutase testis mencit, selanjutnya dianalisis dengan ANOVA dan dilanjutkan dengan uji BNT 1%.
Hasil uji ANOVA menunjukkan bahwa pemberian minyak jintan hitam berpengaruh terhadap kualitas spermatozoa meliputi viabilitas spermatozoa, motilitas spermatozoa, abnormalitas spermatozoa dan kadar SOD testis mencit yang dipapar Pb asetat peroral. Pemberian dosis 1,2 mg/gr/BB minyak jintan hitam adalah dosis yang efektif dalam mempengaruhi kualitas spermatozoa maupun mempengaruhi kadar SOD testis mencit yang dipapar Pbperoral.
ABSTRACT
Lead acetate is one source of ROS that can increase ROS become uncontrolled which can lead to a decreasein male infertility, because it can cause oxidative stress which would cause a decrease in motility, viability and a crosome damage to spermatozoa that can damage the enzyme superoxide-dismutase (SOD), which SOD is in charge of maintaining the function of the cell so that the cell function decline and become damaged. Therefore, the aim of this study is to determine the effect of ethanol extract of black cumin to the quality of spermatozoa and testicular SOD superoxide dismutase levels in mice exposedorally asett Pb.
This research is an experimental research using complete lyrandomized design (CRD) with 4 replications and 5 times treatment. The experimental animals used were mice strain balb / c males aged 2-3 months, for about 20 mice. The treatment in this research include the exposure of lead (Pb) 0.3 mg /grBB / day / orally for 5 days, black cumin ethanol extract at a dose of 0.6 mg /grBB, and a dose of 1.2 mg / grBB for 10 days. The results observed in this research are the quality of sperm and testicular levels of superoxide dismutase in mice, which continue to be analyzed by ANOVA. If there is a very real difference it will be continued with LSD 1%.
The results of ANOVA test showed that the ethanol extract of black cumin affect on sperm quality and testicular SOD in mice which were exposed to lead acetate orally. Dose of 1.2mg / g / BB ethanol extract of black cumin is the dose that is effective in influencing the quality of sperm and affect testicular SOD in mice exposed orally Pb.
