Abstract
ABSTRAK
Lada hitam (Piper nigrum L.) merupakan rempah-rempah yang sering digunakan dikalangan masyarakat dalam setiap masakan. Lada hitam (Piper nigrum L.) berkhasiat sebagai bahan penyegar, penghangat badan, merangsang semangat, obat perut kembung, merangsang keluarnya keringat dan obat sesak nafas. Selain itu, juga sebagai diuretik, dan analgesik. Lada hitam (Piper nigrum L.) mengandung bahan aktif berupa alkaloid piperin yang diduga mampu menekan hormon reproduksi betina sehingga perkembangan folikel ovarium akan terhambat, baik primer, sekunder, tertier maupun de Graff. Namun ada juga yang mengatakan bahwa lada hitam (Piper nigrum L.) bersifat toksik dan berpengaruh terhadap kematian hewan coba. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak n-heksan lada hitam (Piper nigrum L.) terhadap organ reproduksi dan durasi kematian mencit (Mus musculus).
Penelitian dilakukan di Laboratorium Biosistematik Jurusan Biologi Fakultas Saintek Univesitas Islam Negeri Maulana Malik Ibrahim Malang pada bulan Agustus sampai Desember 2012. Penilitian ini bersifat eksperimental menggunakan Rancangan Acak Kelompok (RAK) dengan 4 kali ulangan, apabila terdapat perbedaan nyata maka dilanjutkan dengan uji Beda Nyata Terkecil (BNT) 5%. Perlakuan yang digunakan adalah ekstrak lada hitam (Piper nigrum L.) dengan dosis 25 mg/Kg bb, 50 mg/Kg bb, 75 mg/Kg bb dan 100 mg/Kg bb. Hewan yang digunakan adalah mencit (Mus musculus) betina fertil sebanyak 20 ekor. Data hasil penelitian meliputi jumlah folikel primer, sekunder, tertier dan de Graff.
Hasil penelitian menunjukkan bahwa ekstrak n-heksan lada hitam lada hitam (Piper nigrum L.) dosis 25 mg/Kg bb, 50 mg/Kg bb, 75 mg/Kg bb dan 100 mg/Kg bb tidak berpengaruh terhadap perkembangan folikel primer, sekunder, tertier maupun de Graff. Hal tersebut terjadi diprediksikan pendeknya jangka waktu perlakuan dikarenakan lada hitam (Piper nigrum L.) bersifat toksik pada organ pencernaan, sehingga berakibat kematian.
ABSTRACT
Black pepper (Piper nigrum L.) is a spice commonly used among society in every cuisine. Black pepper (Piper nigrum L.) efficacious as a freshener, body warmers, stimulate enthusiasm, flatulence drugs, drug stimulates perspiration and shortness of breath. In addition, it is also as a diuretics, and analgesics. Black pepper (Piper nigrum L.) contains the active ingredient in the form of alkaloid piperine alleged suppressing female reproductive hormones so that the development of ovarian follicles is inhibited, whether primary, secondary, tertiary and de Graff. But it is also said that the black pepper (Piper nigrum L.) is toxic and affects the animal's death. This study aimed to determine the effect of n-hexane extract of black pepper (Piper nigrum L.) on reproductive organs and death duration of mice (Mus musculus).
The study was conducted at the Biosystematical Laboratory of Biology Departementof Faculty of Science and Technology Maulana Malik Ibrahim State Islamic University of Malang from August to December 2013. The studies use is experimental randomized block design (RAK) with four replications, where if there is a real difference it is followed by least significant difference test (BNT) 5%. The treatment used is an extract of black pepper (Piper nigrum L.) at a dose of 25 mg / kg bb, 50 mg / kg bb, 75 mg / Kg mm and 100 mg / kg bb. Animals used were mice (Mus musculus) as many fertile females 20. Research data include the number of follicles of primary, secondary, tertiary and de Graff.
The results of study shows that the extract of black pepper (Piper nigrum L.) dose of 25 mg / kg bb, 50 mg / kg bb, 75 mg / Kg mm and 100 mg / Kg mm has no effect on follicular development of primary, secondary, tertiary and de Graff . It is predicted to occur due to short-term treatment because black pepper (Piper nigrum L.) is toxic to the digestive organs which results in death
