Isolasi Bakteri Pendegradasi Hidrokarbon sebagai Penghasil Enzim Lipase dan Protease dari Tanah Tercemar Minyak Bumi di Kecamatan Wonocolo Kabupaten Bojonegoro

Indonesia merupakan salah satu negara yang kaya akan sumber daya alam baik di darat maupun di laut. Salah satu contoh sumber daya alam yang terdapat di Indonesia adalah pertambangan minyak bumi di Kecamatan Wonocolo Kabupaten Bojonegoro. Adanya aktivitas pengeboran mengakibatkan tanah dilokasi pertambangan tercemar, sehingga menimbulkan kerusakan ekosistem dan mikroorganisme yang terdapat pada tanah tersebut.
Tujuan penelitian ini yaitu untuk mendapatkan bakteri yang berpotensi mendegradasi hidrokarbon sebagai penghasil enzim lipase dan protease. Metode penelitian yang digunakan yaitu jenis penelitian deskriptif eksploratif untuk memberi gambaran mengenai bakteri yang berasal dari tanah tercemar minyak bumi di Kecamatan Wonocolo, Kabupaten Bojonegoro. Jenis deskriptif eksploratif ini meliputi keberadaan bakteri pendegradasi hidrokarbon yang terdapat pada tanah tercemar minyak bumi yang meliputi karakteristik makroskopik, mikroskopis analisis uji biokimia, potensi bakteri penghasil enzim lipase dan protease.
Hasil penelitian menunjukkan bahwa terdapat 5 isolat bakteri dengan kemampuan 1 isolat terpilih yaitu TPY 1 yang diduga sebagai bakteri pendegradasi hidrokarbon penghasil enzim lipase dan protease.

Indonesia is a country rich in natural resources both on land and at sea. One example of natural resources found in Indonesia is petroleum mining in Wonocolo District, Bojonegoro Regency. The existence of drilling activities resulted in polluted soil at the mining location, causing damage to the ecosystem and microorganisms contained in the soil.
The purpose of this study was to obtain bacteria that have the potential to degrade hydrocarbons as a producer of lipase and protease enzymes. The research method used is descriptive exploratory research to provide an overview of bacteria originating from oil-contaminated soil in Wonocolo District, Bojonegoro Regency. This exploratory descriptive type includes the presence of hydrocarbon-degrading bacteria found in oil-contaminated soil which includes macroscopic characteristics, microscopic analysis of biochemical tests, the potential of bacteria producing lipase and protease enzymes.
The results showed that there were 5 bacterial isolates with the ability of 1 selected isolate, namely TPY 1 which was suspected as a hydrocarbon-degrading bacteria producing lipase and protease enzymes.

إندونيسيا بلد غني بالموارد الطبيعية في البر والبحر. أحد الأمثلة على الموارد الطبيعية الموجودة في إندونيسيا هو تعدين البترول في مقاطعة وونوكولو ، بوجونيغورو ريجنسي. أدى وجود أنشطة الحفر إلى تلوث التربة في موقع التعدين ، مما تسبب في تلف النظام البيئي والكائنات الحية الدقيقة الموجودة في التربة. كان الغرض من هذه الدراسة هو الحصول على البكتيريا التي لديها القدرة على تحلل الهيدروكربونات كمنتج للليباز وإنزيمات البروتياز
طريقة البحث المستخدمة هي البحث الاستكشافي الوصفي لتقديم لمحة عامة عن البكتيريا التي تنشأ من التربة الملوثة بالزيت في منطقة ونوكولو ، بوجونيغورو ريجنسي. يشتمل هذا النوع الوصفي الاستكشافي على وجود بكتيريا مهينة للهيدروكربون الموجودة في التربة الملوثة بالزيت والتي تشمل الخصائص العيانية والتحليل المجهري للاختبارات الكيميائية الحيوية وإمكانية إنتاج البكتيريا للليباز وإنزيمات البروتياز.
. أظهرت النتائج وجود ٥ عزلات بكتيرية ذات قدرة عزلة واحدة مختارة وهي TPY ١ والتي تم الاشتباه في أنها بكتريا مهينة للهيدروكربونات منتجة للليباز وإنزيمات البروتياز.
