Abstract
ABSTRAK
Infeksi Saluran Kemih (ISK) merupakan salah satu penyakit infeksi tersering di dunia yang disebabkan oleh bakteri Escherichia coli. Pengobatan penyakit ini dengan menggunakan antibiotik, namun bakteri Escherichia coli saat ini sudah mulai resisten dengan berbagai antibiotik, sehingga diperlukan adanya inovasi baru bahan yang dapat digunakan sebagai antibiotik terhadap Escherichia coli. Penelitian ini bertujuan untuk mengetahui kemampuan Aspergillus niger pada konsentrasi tertentu sebagai antibakteri dan antibiofilm terhadap Escherichia coli dengan melakukan uji Kadar Hambat Minimum (KHM), uji Kadar Bunuh Minimum (KBM), dan uji Minimum Biofilm Inhibitory Concentration (MBIC) yang dilakukan pada well-microplate. Pada uji KHM dan uji MBIC dilakukan pembacaan nilai optical density menggunakan microplate reader dan didapatkan hasil nilai terendah pada konsentrasi 25% sebesar 0.388 dan 0.587. Sedangkan pada uji KBM diamati pada dasar microplate dan ditemukan adanya endapan pada dasar microplate. Sehingga dari hasil penelitian dapat disimpulkan nilai KHM dan MBIC berada pada konsentrasi 25% berturut-turut adalah 46% dan 56%, sedangkan pada uji KBM didapatkan hasil Aspergillus niger tidak mampu membunuh bakteri Escherichia coli.
ABTRACT
Urinary Tract Infection (UTI) is one of the most common infectious diseases in the world caused by Escherichia coli bacteria. The treatment of this disease is by using antibiotics, but the Escherichia coli bacteria are now starting to become resistant to various antibiotics, so that new material innovations are needed that can be used as antibiotics against Escherichia coli. This study aims to determine the ability of Aspergillus niger at certain concentrations as an antibacterial and antibiofilm against Escherichia coli by performing the Minimum Inhibitory Concentration (MIC) test, Minimum Killing Concentration (MBC) test, and Minimum Biofilm Inhibitory Concentration (MBIC) test which was carried out on wells. microplate. In the MIC test and MBIC test, the optical density value was read using a microplate reader and the lowest values were obtained at a concentration of 25% of 0.388 and 0.587. Meanwhile, the KBM test was observed on the bottom of the microplate and found deposits on the bottom of the microplate. So from the results of the study it can be concluded that the MIC and MBIC values are at a concentration of 25%, 46% and 56%, respectively, while the MBC test shows that Aspergillus niger is unable to kill Escherichia coli bacteria.
