Abstract
INDONESIA:
Diperkirakan sebanyak 75% wanita di Indonesia pernah mengalami keputihan. Keputihan paling sering diakibatkan oleh infeksi jamur Candida albicans dan dapat menyebabkan komplikasi yang serius contohnya infertilitas. Salah satu bahan ramuan yang terkenal dapat menyembuhkan masalah-masalah infertilitas adalah rimpang jeringau (Acorus calamus L.). Penelitian ini menjadi langkah awal untuk proses standardisasi dan saintifikasi rimpang jeringau sebagai salah satu bahan dasar obat tradisional etnis Madura, yaitu jamu “Subur Kandungan”. Sehingga dilakukan penelitian lebih mendalam sebagai antioksidan dan antifungi dengan beberapa pelarut organik berdasarkan tingkat kepolaran.
Penelitian ini menggunakan penelitian experimental design. Sampel diekstrak dengan metode maserasi menggunakan pelarut etanol p.a, kloroform p.a, dan n-heksana p.a. Uji kadar antioksidan menggunakan variasi konsentrasi 25 ppm, 50 ppm, 100 ppm, 200 ppm, dan 400 ppm. Kemudian dihitung persen aktivitas antioksidannya. Selanjutnya diuji aktivitas antifungi secara in vitro terhadap pertumbuhan jamur Candida albicans dengan konsentrasi 100%; 50%; 25%; 12,5%; 6,25%; 3,13%; 1,56%; 0,78%; 0,39%; dan 0%. Pada masing-masing perlakuan dilakukan ulangan sebanyak tiga kali.
Hasil nilai IC50 uji aktivitas antioksidan ekstrak kasar rimpang jeringau dengan metode DPPH dari tertinggi ke terendah berturut-turut (etanol; 137,7 mg/L tergolong sedang), (kloroform; 315,8 mg/L tergolong lemah), dan (n-heksana; 1011 mg/L tergolong sangat lemah/tidak aktif). Sedangkan zona hambat dengan diameter dari ukuran terbesar sampai terkecil secara berurutan adalah kontrol positif (nystatin) 17,68 mm (kuat), jeringau etanol 3,72 mm (sedang), jeringau n-heksana 3,32 mm (sedang), jeringau kloroform 2,2 mm (lemah) dan kontrol negatif (pelarut etanol 70%) 0,77 mm (lemah). Adapun nilai KHM terdapat pada masing-masing ekstrak rimpang jeringau konsentrasi 0,39%. Dan nilai KBM ekstrak rimpang jeringau masing-masing perlakuan didapatkan pada konsentrasi 0,78%.

It is estimated that around 75% of women in Indonesia have experienced vaginal discharge. Vaginal discharge is most frequently caused by the infection of Candida albicans and can cause serious complications such as infertility. One well-known concoction that can cure infertility problems are Jeringau rhizomes (Acorus Calamus L.). This research is the first step to the process of standardization and scientification of Jeringau rhizome as a basic ingredient of traditional medicine used by Madurese ethnic, namely “Subur Kandungan”. Hence, the more in-depth research is also conducted using numerous organic solvents in extraction process.
This research is an experimental design research. Sample was extracted using maceration method with solvents of ethanol p.a, chloroform p.a, dan n-hexane p.a. Examine the antioxidant levels using a variation of the concentration—25 ppm, 50 ppm, 100 ppm, 200 ppm and 400 ppm. Then, the researcher calculated the percentage of antioxidant activity. The antifungal activity in vitro against Candida albicans fungal growth with a concentration of 100%; 50%; 25%; 12.5%; 6.25%; 3.13%; 1.56%; 0.78%; 0.39%; and 0% is examined further. Each treatment is conducted three times.
The IC50 values test in the experiment related to the antioxidant activity of Jeringau rhizomes crude extract using DPPH from highest to lowest are as follows: (ethanol; 137.7 mg/L was moderate), (chloroform; 315.8 mg/L relatively weak), and (n-hexane; 1011 mg/L as very weak/inactive). Whereas the inhibition zones with a diameter the size from the largest to the smallest are a positive control (nystatin)—17.68 mm (strong)—, Jeringau ethanol—3 .72 mm (medium)—, Jeringau n-hexane—3.32 mm (medium)—, Jeringau chloroform—2.2 mm (weak)—, and negative controls (solvent ethanol 70%)—0.77 mm (weak)—. Therefore, it can be concluded that the MIC values contained in each extract of Jeringau rhizomes is 0.39% and the MFC values of Jeringau rhizomes extract in each treatment is obtained at the concentration of 0.78%.
