Abstract
ABSTRACT
HMG CoA reductase is a receptor that plays an important role in cholesterol formation. The receptor can bind to HMG CoA so that it can form cholesterol. When genetic mutations occur, it can cause excessive cholesterol production which can cause hypercholesterolemia. Hypercholesterolemia can be prevented by using squalene which has anti-cholesterol potential. Kosambi (Scleichera oleosa) is a plant that contains squalene composition. This research discusses kosambi (Schleichera oleosa) which can help the formation of cholesterol In Silico. The type of research conducted is exploratory descriptive. Ligand preparation, receptor protein preparation, Human Intestinal Absorption test (HIA), Lippinski Rule of five (Ro5) Test, Prediction of Activity Spectra for Substances (PASS), Molecular Docking (Molecular Docking) and visualization of docking results. Data were analyzed and compared with simvastatin as a comparison based on the category of Human Intestine Absorption (HIA), Lippinski Rule of five (Ro5), and the likelihood value of the activity (Pa) from the Prediction of Activity Spectra for Substances (PASS). The docking results are developed by looking at the best model that has the lowest energy bond and type bond formed. Schlesichera oleosa tested has the best binding value of affinity of -4.9 kcal/ mole and has been shown to have anti-hypercholesterolemia activity. Squalene also has a good ability to be absorbed in the intestine and can penetrate cells so that the approved data can be an anti-cholesterol candidate.

HMG CoA reductase merupakan reseptor yang berperan penting dalam pembentukan kolesterol. Reseptor tersebut dapat berikatan dengan HMG CoA sehingga dapat membentuk kolesterol. Ketika terjadi mutasi genetik dapat menadikan produksi kolesterol yang berlebihan sehingga dapat memicu hiperkoletserolemia. Hiperkolesterolemia dapat dicegah menggunakan senyawa squalene yang memiliki potensi antikolesterol. Kosambi (Scleichera oleosa) merupakan salah satu tanaman yang mengandung senyawa squalene. Penelitian ini bertujuan untuk mengetahui bahwa kosambi (Schleichera oleosa) dapat menghambat pembentukan kolesterol secara In Silico. Jenis penelitian yang dilakukan adalh deskriptif eksploratif. Penelitian ini dilakukan dengan beberapa tahapan meliputi: preparasi ligan, preparasi protein reseptor, uji Human Intestinal Absorption (HIA), Uji Lippinski Rule of five (Ro5), Uji Prediction of Activity Spectra for Substances (PASS), penambatan molekular (Molecular Docking) dan visualisasi hasil docking. Data dianalisis dan dibandingkan dengan simvastatin sebagai pembanding berdasarkan kategori Human Intestinal Absorption (HIA), Lippinski Rule of five (Ro5) dan nilai Probable Activity (Pa) dari Uji Prediction of Activity Spectra for Substances (PASS). Hasil docking dianalisis dengan melihat model terbaik yang memiliki energi ikatan paling rendah dan jenis ikatan terbentuk. Hasil penelitian menunjukkan bahwa senyawa squalene kosambi (Schleichera oleosa) yang diujikan memiliki nilai binding affinity terbaik yaitu -4,9 kkal/mol dan terbukti memiliki aktivitas sedang sebagai antihiperkolesterolemia. Squalene juga memiliki kemampuan terabsorbsi baik di usus dan mampu menembus membrane sel sehingga data direkomendasikan sebagai senyawa yang dapat menjadi kandidat antikolesterol.
