Abstract
ABSTRACT
Pneumonia is a health problem with high mortality, especially Ventilator-Associated Pneumonia (VAP). The most common cause of VAP is Klebsiella pneumoniae. This bacterium can make strong biofilms that are difficult to eradicate. The black mulberry leaf contains tannin, terpenoids, steroids and anthocyanins which are believed to have an antibiofilm effect on K. pneumoniae. This study aims to study the activity of black mulberry leaf extract as attachment prevention, growth inhibition and destruction of K. pneumoniae biofilms. The method used is the tissue culture plate method. The treatments consisted of black mulberry leaf extract with a concentration 0.16 mg/ml, 0.08 mg/ml, 0.04 mg/ml, 0.02 mg/ml, and 0.01 mg/ml. The results of the study were read using a microplate reader and optical density (OD) values were obtained. The results showed that black mulberry leaves had activity against attachment, growth inhibition, and destruction of K. pneumoniae biofilms with the highest activity at concentrations 0.04 mg/ml, 0.04 mg/ml, and 0.16 mg/ml. The percentage of antibiofilm activity in these combinations was 28.13%, 19.68%, and 25.70%. The results of the analysis data on the attachment prevention and growth inhibition of biofilm showed that the value of optical density was not influenced by the large concentration of black mulberry leaf extract. Meanwhile, in the biofilm destruction test, the value of optical density was significantly affected by the large concentration of black mulberry leaf extract (Morus nigra L.).
ABSTRAK
Pneumonia merupakan masalah kesehatan dengan angka kematian yang tinggi, terutama Ventilator Associated Pneumonia (VAP). Penyebab terbanyak VAP adalah Klebsiella pneumoniae. Bakteri ini memiliki kemampuan membentuk biofilm kuat sehingga sulit untuk dieradikasi. Daun murbei hitam memiliki kandungan senyawa tannin, terpenoid, steroid dan antosianin yang dipercaya memiliki efek antibiofilm terhadap K. pneumoniae. Penelitian ini bertujuan untuk mengetahui aktivitas ekstrak daun murbei hitam sebagai pencegah perlekatan, penghambat pertumbuhan dan penghancuran biofilm K. pneumoniae. Metode yang digunakan adalah metode tissue culture plate. Perlakuan berupa penambahan ekstrak daun murbei hitam dengan konsentrasi 0,16 mg/ml, 0,08 mg/ml, 0,04 mg/ml, 0,02 mg/ml dan 0,01 mg/ml. Hasil penelitian dibaca menggunakan microplate reader dan diperoleh nilai optical density (OD). Hasil penelitian menunjukkan bahwa daun murbei hitam memiliki aktivitas pencegahan perlekatan, penghambatan pertumbuhan, dan penghancuran biofilm K.
pneumoniae dengan aktivitas tertinggi pada konsentrasi berturut-turut yaitu 0,04 mg/ml, 0,04 mg/ml, dan 0,016 mg/ml. Presentase aktivitas antibiofilm pada konsentrasi tersebut berturut-turut yaitu 28,13%, 19,68% dan 25,70%. Hasil analisis data penelitian pada uji pencegahan perlekatan dan penghambatan pertumbuhan biofilm menunjukkan bahwa besar nilai optical density tidak dipengaruhi oleh besar konsentrasi ektrak daun murbei hitam. Sedangkan, pada uji penghancuran biofilm besar nilai optical density dipengaruhi secara signifikan oleh besar konsentrasi ektrak daun murbei hitam (Morus nigra L.).
