Abstract
INDONESIA:
Enzim Protease dapat diproduksi oleh berbagai jenis mikroba. Pada umumnya oleh jenis bakteri dan kapang proteolitik. Bakteri proteolitik dapat diperoleh dari bekatul yang memiliki kadar protein yang cukup tinggi. Tujuan penelitian ini untuk mengisolasi bakteri proteolitik dari bekatul dan menguji aktivitas protease hasil isolat yang dihasilkan.
Metode penelitian ini meliputi isolasi dan seleksi bakteri proteolitik dari bekatulselanjutnya diidentifikasi secara makroskopis dan mikroskopis sampai identifikasi spesies. Hasil isolat selanjutnya dilakukan uji aktivitas enzim protease pada berbagai suhu.
Hasil penelitian ini menunjukkan bahwa bakteri proteolitik yang diisolasi dari bekatul didapatkan sebanyak tiga isolat yaitu isolat J, K, dan L. Uji makroskopis menunjukkan bahwa ke 3 isolat J berbentuk bulat kecil, elevesi tidak rata, tepian bergerigi, dan warna krem. Isolat K berbentuk bulat kecil, elevesi rata, tepian rata, dan warna krem. Sedangkan isolat L berbentuk bulat besar, elevesi cembung, tepian rata, dan warna krem.Pada uji mikroskopis yang meliputi uji katalase dan pewarnaan Gram, menunjukkan hasil yang positif dari ketiga isolate tersebut. Pada identifikasi spesies yang menggunakan microbact mengahasilkan jenis bakteri berupa Bacillus firmus. Ketiga isolat tersebut didapatkan isolat terbaik dengan indeks proteolitik (IP) terbesar 7,6 yaitu isolat K. Aktivitas protease yang dihasilkan isolat K optimum padasuhu 30 ºC sebesar 0,7203 Unit/mL.

Protease enzymes can be produced by various types of microbes. In general, by the type of bacteria and proteolytic mold. Proteolytic bacteria can be obtained from rice bran which has a high enough protein. The purpose of this study was to isolate proteolytic bacteria from rice bran and test the protease activity of the resulting isolates.
This research method includes isolation and selection of proteolytic bacteria from rice bran then macroscopically and microscopically identified to species identification. Results of the isolate tested protease enzyme activity was from the at various temperature.
The results of this study showed that proteolytic bacteria isolated from rice bran were obtained as many as three isolates, namely isolates J, K, and L. Macroscopic tests showed that the 3 isolates of J were small rounded, uneven elevesi, jagged edges, and cream color. K isolates are small, round elevesi, flat edges and cream colors. While the L isolates were large, convex elves, flat edges and cream colors. Microscopic tests which included catalase tests and Gram staining showed positive results from the three isolates. In the identification of species that use microbacts produced bacteria in the form of Bacillus firmus. The three isolates obtained the best isolates with the largest Proteolytic Index of 7.6, isolate K. The protease activity produced by the optimum K isolate at a temperature of 30 ºC was 0.7203 Unit / mL
