Abstract
INDONESIA:
Tanaman anting-anting (Acalypha indica L.) merupakan salah satu tumbuhan yang dapat digunakan sebagai obat herbal, karena di dalamnya mengandung metabolit sekunder. Salah satu metabolit sekunder yang memiliki sifat farmakologi adalah alkaloid. Alkaloid yang terdapat pada tanaman anting-anting (Acalypha indica L.) diuji kestabilannya untuk melihat ada tidaknya penurunan intensitas warna secara visual selama proses analisis, dan perubahan nilai Rf berdasarkan perbedaan kelembaban pada proses pemisahan.
Pengambilan metabolit sekunder dalam sampel dilakukan dengan ekstraksi ultrasonik menggunakan pelarut etil asetat. Kestabilan visual alkaloid dilakukan menggunakan metode KLT dengan memvariasikan waktu pengamatan UV dengan selang waktu 0, 5, 10, 20, 30, dan 60 menit. Sedangkan, identifikasi perubahan nilai Rf dilakukan elusi pada pagi dan siang hari dengan mengukur kelembaban ruang.
Hasil penelitian menujukan bahwa tidak adanya noda alkaloid pada pelat KLT yang memudar atau mengalami penurunan intensitas warna selama 60 menit pengamatan. Perbedaan kelembaban ruang tidak mempengaruhi nilai Rf pada pelat KLT dengan nilai standar deviasi (SD) yaitu ≤ 0,01. Tidak berpengaruhnya kelembaban terhadap nilai Rf dikarenakan perbedaan kelembaban pagi dan siang hari yang sangat sedikit.

Anting-anting (Acalypha indica L.) is one of a plant that can be used as a herbal medicine, because it contains secondary metabolites. One of secondary metabolites that has pharmacological effect is an alkaloid. Alkaloids that contains in Anting-anting (Acalypha indica L.) was tested for stability to see whether there was decrease in color intensity visually during the analysis process, and changes Rf values based on differences in humidity in the separation process.
Intake of secondary metabolites in the sample was carried out by ultrasonic extraction using ethyl acetate solvent. Alkaloid visual stability using TLC method by varying the time of UV visualization with intervals of 0, 5, 10, 20, 30, and 60 minutes. While, the identification of changes Rf values carried out elution in the morning and afternoon by measuring the humidity of the room.
The results showed that there were no spots of alkaloid on the TLC plates that faded or decreased color intensity during 60 minutes of observation. The difference in room humidity does not affect the Rf values on the TLC plate with a standard deviation (SD) value that is ≤ 0.01. Humidity does not affect Rf value, due to very little difference between morning and afternoon humidity.
