Abstract
INDONESIA :
Senyawa steroid dan triterpenoid diisolasi dari mikroalga Chlorella sp. menggunakan kromatografi kolom dengan variasi diameter kolom. Chlorella sp. dikultivasi dalam Medium Ekstrak Tauge (4%) dan dipanen pada hari kesepuluh menggunakan metode sentrifugasi. Tahapan isolasi steroid dan triterpenoid mikroalga Chlorella sp. berupa ekstraksi maserasi menggunakan pelarut metanol p.a, hidrolisis menggunakan HCl 2 N, partisi dengan petroleum eter dan pemisahan menggunakan kromatografi kolom. Hasil rendemen ekstraksi maserasi sebesar 23,79% dan rendemen partisi sebesar 55,06%. Variasi diameter kolom yang digunakan adalah 1 cm, 1,5 cm dan 2 cm. Hasil pemisahan dimonitoring menggunakan KLT. Hasil monitoring menunjukkan kolom berdiameter 1 cm menghasilkan 1 fraksi steroid (Rf 0,69) dan 2 fraksi triterpenoid (Rf 0,50 dan 0,33). Kolom berdiameter 1,5 cm menghasilkan 1 fraksi steroid (Rf 0,44) dan 3 fraksi triterpenoid (Rf 0,30; 0,21 dan 0,14). Kolom diameter 2 cm menghasilkan fraksi terbanyak dengan 2 fraksi steroid (Rf 0,94 dan 0,56) dan 3 fraksi triterpenoid (Rf 0,44; 0,26 dan 0,19).
ENGLISH :
Steroid and triterpenoid compounds were isolated from microalgae Chlorella sp. using chromatography column with the varied diameter column. Chlorella sp. was cultivated in a Medium Extract of Sprout (4%) and harvested in the 10th day applying a centrifuge. There are several steps to isolate steroid and triterpenoid. They are macerations with methanol p.a, hydrolysis using HCl 2N, partition with petroleum ether and separation using column chromatography. The yield of maceration was 23.79% while the partition yield was 55.06%. The variations of column diameter used in this research were 1 cm, 1.5 cm, and 2 cm. The separation products were monitored by the use of TLC. The result of monitoring by TLC using diameter column 1 cm showed that there were 1 fraction of steroid (Rf 0.69) and 2 fractions of triterpenoid (Rf 0.50 and 0.33). The separation using diameter column 1.5 cm obtained 1 steroid fraction (Rf 0.44) and 3 triterpenoid fractions (Rf 0.30, 0.21, and 0.14). The best separation was shown in diameter column 2 cm obtaining 2 steroid fractions (Rf 0.94 and 0.56) and 3 triterpenoid fractions (Rf 0.44, 0.26, and 0.19).
