Pengaruh pemberian ekstrak etanol 70% biji rambutan (Nephelium lappaceum L.) terhadap kualitas sperma mencit (Mus muscullus L.) yang diinduksi streptozotocin

Rambutan (Nephelium lappaceum L.) merupakan tanaman yang mempunyai manfaat sebagai obat herbal. Biji tanaman ini mempunyai manfaat sebagai antidiabetik. Biji rambutan mempunyai senyawa flavonoid yang dapat mencegah kerusakan dan meningkatkan sekresi insulin pada sel β sehingga mampu memberikan efek hipoglikemik. Kondisi penderita diabetes mengalami penurunan kualitas sperma. Kandungan flavonoid dari biji rambutan diduga mampu memperbaiki dan meningkatkan kualitas sperma. Tujuan penelitian ini untuk mengetahui pengaruh pemberian ekstrak etanol 70% biji rambutan terhadap kualitas sperma mencit diabetes.

Penelitian ini termasuk penelitian eksperimental yang termasuk Rancangan Acak Lengkap (RAL) dengan 6 perlakuan dan 4 ulangan, mencakup kelompok K- (mencit sehat), K+ (metformin 39 mg/kgBB), perlakuan penambahan ekstrak P0 (CMC 0,5%), P1 (15 mg/kgBB), P2 (19,2 mg/kgBB), P3 (23,4 mg/kg BB). Parameter kualitas sperma yang diamati mencakup viabilitas, abnormalitas, motilitas pada sperma. Data hasil penelitian dianalisis menggunakan One Way Anova dan dilakukan uji lanjut menggunakan uji Duncan 5%.

Berdasarkan hasil Uji Duncan memperlihatkan adanya perbedaan nyata dari setiap perlakuan ekstrak etanol 70% biji rambutan (Nephelium lappaceum L.). Parameter abnormalitas sperma menunjukkan dosis 23,4 mg/kgBB sebagai dosis yang efektif, sedangkan parameter viabilitas dan motilitas sperma dosis efektif menggunakan 19,2 mg/kgBB. Ekstrak etanol 70% biji rambutan (Nephelium lappaceum L.) terbukti dapat meningkatkan kualitas sperma mencit (Mus muscullus L.) yang diinduksi streptozotocin.

Rambutan (Nephelium lappaceum L.) is a plant that has been known as an herbal medicine. Seed of this plant have benefits as an antidiabetic. Rambutan seeds have flavonoid compounds that can prevent damage and increase insulin secretion on β cells so as to provide a hypoglycemic effect. The condition of sperm also decreased quality caused of diabetic condition. The content of flavonoids is known to improve sperm quality. The purpose of this research is to know the potency of rambutan seed 70% ethanol extract on the sperm quality of diabetic mice.

This research is an experimental research using Completely Randomized Design with 6 treatments and 4 replications, including the group of K- (healthy mice), K + (metformin 39 mg/kgBW), treatment of addition of P0 extract (CMC 0.5%), P1 (15 mg/kgBW), P2 (19.2 mg/kgBW), P3 (23.4 mg/kgBW). Sperm quality parameters were observed about viability, abnormality and motility. Data of the research were analyzed using One Way Anova and further testing using 5% Duncan test.

Based on the Duncan Test results showed a real difference from each treatment of rambutan seed (Nephelium lappaceum L.) 70% ethanol extract. The dose of 23.4 mg/kgBW showed as the most effective dose of sperm abnormality parameters, while the parameter of viability and motility of sperm dose effectively use 19.2 mg/ mg/kgBW. Rambutan Seed (Nephelium lappaceum L.) 70% ethanol extract has been shown to improve sperm quality of mice (Mus muscullus L.) induced by streptozotocin.
