Abstract
INDONESIA:
Tujuan penelitian ini untuk mengetahui pengaruh jenis media dan konsentrasi BAP serta interaksi terhadap multiplikasi tunas stevia (Stevia rebaudiana) secara In Vitro. Penelitian ini bersifat eksperimental, menggunakan rancangan acak lengkap (RAL) dengan 12 perlakuan dan 3 ulangan. Perlakuan dalam penelitian ini ada dua faktor yaitu: jenis media meliputi media MS, media DKW dan media WV5 dan konsentrasi BAP meliputi 0,0 µM, 2,2 µM, 4,4 µM dan 6,6 µM. Data dianalisis dengan Uji ANAVA Two-Way α = 5%. Apabila terdapat perbedaan signifikan maka dilanjutkan uji Duncan Multiple Range Test (DMRT) dengan taraf signifikan 5%. Hasil dari penelitian menunjukkan media terbaik untuk perbanyakan tunas adalah media WV5. Konsentrasi terbaik untuk perbanyakan tunas pada BAP 4,4 µM. Hari mucul tunas tercepat pada MS + BAP 4,4 µM dan WV5 + BAP 6,6 µM. Hari muncul daun tercepat pada DKW + BAP 2,2 µM. Hari daun membuka sempurna tercepat pada DKW + BAP 0,0 µM. jumlah daun dan tinggi planlet terbaik pada MS + BAP 0,0 µM. dan berat basah terbaik pada perlakuan DKW + BAP 2,2 µM.
ENGLISH:
The purpose of this study was to determine the effect of media type and concentration of BAP and the interaction on the multiplication of stevia buds (Stevia Rebaudiana) in vitro method. This study was experimental, using a completely randomized design (CRD) with 12 treatments and 3 replications. The treatment in this study there were two factors: media type including MS media, DKW media and WV5 media and BAP concentrations included 0.0 µM, 2.2 µM, 4.4 µM and 6.6 µM. Data were analyzed by ANAVA Test Two-Way α = 5%. If there are significant differences then continued by the Duncan Multiple Range Test (DMRT) with a significant level of 5%. The results of the study showed the presence of the influence of media types and concentrations of BAP against multiplication shoots stevia. Results showed that the best medium on multiplication shoots of MS. The best concentration on multiplication shoots of BAP 4,4 µM. The day appeared the fastest shoots from the medium MS + BAP 4,4 µM and WV5 + BAP 6,6 µM. The day appeared the fastest leaves from the medium DKW + BAP 2,2 µM. The day leaves open the perfect from the medium DKW + BAP 0,0 µM. The number of Shoots and the highplanlet from medium MS + BAP 0,0 µM. and the best wet weight for treatment DKW + BAP 2,2 µM.
