Abstract
INDONESIA:
Isolasi BAL dari lingkungan alam telah banyak dilakukan pada buah-buahan seperti pada buah mangga manalagi (Mangifera indica). Keberadaan bakteri probiotik (BAL pada buah mangga manalagi) dapat mencegah pertumbuhan bakteri patogen seperti Escherichia coli dan Staphylococcus aureus. Escherichia coli merupakan penyebab penyakit diare sedangkan Staphylococcus aureus merupakan bakteri patogen oportunistik pada manusia yang menyebabkan diare, infeksi kulit, bakteremia, endokarditis, osteomielitis.
Penelitian dilakukan dengan rancangan kualitatif dan kuantitatif meliputi pengamatan mikroskopis, makroskopis, dan uji biokimia (uji katalase, uji motilitas, uji tipe fermentasi, uji pertumbuhan bakteri pada suhu yang berbeda, dan uji pertumbuhan bakteri pada konsentrasi NaCl yang berbeda) dari masing-masing isolat BAL buah mangga manalagi, serta pengujian isolat BAL yang mampu menghasilkan antibakteri Staphylococcus aureus dan Escherichia coli menggunakan metode sumuran.
Hasil pada pengujian makroskopis, mikroskopis dan biokimiawi dari isolat bakteri asam laktat pada buah mangga manalagi (Mangifera indica) termasuk genus Streptococcus, genus Enterococcus, dan tiga isolat BAL yang termasuk genus Lactobacillus. Serta kemampuan isolat BAL buah mangga manalagi yang merupakan genus Lactobacillus, Streptococcus dan Enterococcus berpotensi kuat menghasilkan antibakteri Escherichia coli dan Staphylococcus aureus.
ENGLISH:
LAB (Lactic Acid Bacteria) isolation from natural environment has frequently carried out on fruit such as Manalagi mango (Mangifera indica). The existence of probiotic bacteria (LAB on Manalagi mangoes) could prevent pathogenic bacteria from growing, especially Escherichia coli and Staphylococcus aureus. The Escherichia coli is a common cause of diarrhea while Staphylococcus aureus is an opportunistic pathogenic bacterium in human which causes diarrhea, skin infections, bacteremia, endocarditis, osteomyelitis.
This research uses qualitative and quantitative designs including microscopic, macroscopic, and biochemical tests (catalases test, motility test, fermentation type test, bacterial growth test at different temperatures, and bacterial growth test at different NaCl concentrations) of each Manalagi mango LAB isolate, as well as testing of LAB isolate which able to produce antibacterial of Staphylococcus aureus and Escherichia coli using the well method.
The results of macroscopic, microscopic and biochemical testing are the lactic acid bacterial isolates on Manalagi mango (Mangifera indica) including Streptococcus genus, Enterococcus genus, and three isolates of LAB belong to Lactobacillus genus. As well as the ability of Manalagi mango LAB isolates from Lactobacillus, Streptococcus and Enterococcus genus which has a potential to produce antibacterial of Escherichia coli and Staphylococcus aureus.
