Abstract
INDONESIA:
Koleksi plasma nutfah C. reticulata L. di Kebun Percobaan Punten milik Balai Penelitian Tanaman Jeruk dan Buah Subtropika (Balitjestro) Batu belum dikarakterisasi secara menyeluruh. Karakterisasi C. reticulata L. selama ini masih dilakukan secara sederhana, yaitu dengan karakterisasi morfologi. Karakterisasi morfologi merupakan teknik yang mudah dan cepat, akan tetapi, teknik karakterisasi tersebut memiliki keterbatasan yaitu dipengaruhi oleh lingkungan, sehingga akan memberikan hasil yang berbeda-beda bergantung pada tempat tumbuhnya. Keterbatasan dari markah morfologi dapat diatasi dengan menggunakan markah molekuler yang berbasis DNA. Salah satu teknik molekuler yang dapat digunakan adalah metode Inter Simple Sequence Repeat (ISSR). Penelitian ini bertujuan untuk mengetahui keragaman genetik 45 aksesi jeruk keprok (Citrus reticulata L.) berdasarkan karakter morfologi dan molekuler ISSR serta korelasi antara karakter morfologi dan molekuler.
Penelitian morfologi dilakukan di Kebun Percobaan Punten milik Balitjestro, Batu. Data morfologi diuraikan secara deskriptif sesuai buku panduan Descriptors for Citrus (IPGRI 1999). dan dibuat dendogram. Sedangkan analisis karakterisasi molekuler dilakukan di Laboratorium Terpadu. Balitjestro. Data pola pita ISSR dianalisis secara kuantitatif berdasarkan muncul tidaknya pita pada gel dan dibuat dendogram. Hubungan kekerabatan antara karakterisasi morfologi dan molekuler dianalisis menggunakan koefisien kemiripan Dice. Korelasi antara keduanya dianalisis menggunakan statistik Z Mantel dengan prosedur MXCOMP pada program NTSYS.
Hasil penelitian menunjukkan hubungan kekerabatan berdasarkan karakterisasi morfologi mempunyai nilai koefisien kemiripan genetik antara 0.41 sampai 1.00 dan diperoleh nilai goodness of fit sebesar 0.84 % (baik), sedangkan nilai koefisien kemiripan genetik pada karakterisasi molekuler didapatkan hasil antara 0.76 sampai 1.00, dan diperoleh goodness of fit sebesar 0.91 % (sangat baik). Nilai korelasi antara karakter morfologi dan molekuler ISSR didapatkan nilai r sebesar -0.00840 P= < 0.321 (sangat lemah). Sehingga dapat disimpulkan bahwa hasil korelasi pengelompokan 45 aksesi C. reticulata berdasarkan karakterisasi morfologi tidak konsisten dengan hasil yang diperoleh dari analisis pola pita DNA, sehingga matriks kemiripan yang berasal dari data morfologi tidak dapat digunakan untuk menduga kemiripan genetik.

The Collection of Nutfah of plasm (C. reticulata L.) in Punten Experimental Garden for Citrus and Subtropical Research Institute (Balitjestro) Batu has not been fully characterized. The Characterization of C. reticulata L. has been done in simple terms, namely morphological characterization. Morphological characterization is an easy and fast technique; however, the characterization technique has a limitation that is influenced by the environment, so it will give different results depending on the growth. Limitations of morphological markers can be resulted by using molecular markers based on DNA. One of the molecular techniques is the method of Inter Simple Sequence Repeat (ISSR). The research aims at determining the genetic diversity of 45 of accessions of tangerine orange (Citrus reticulata L.) based on the morphological and molecular characters of ISSR and the correlation between morphological and molecular characters.
The morphology study was conducted at Balitjestro Punten Experimental Garden, Batu. Morphological data were described descriptively according to the guide book of the Descriptors for Citrus (IPGRI 1999) and made dendogram. The molecular characterization analysis was done in Laboratory of Balitjestro. The ISSR band pattern data was analyzed quantitatively based on the appearance of bands on the gel and it was made dendogram. The kinship relationship between morphological and molecular characterization was analyzed using Dice's resemblance coefficient of. The correlations of both were analyzed using Z Mantel statistics with the MXCOMP procedure on the NTSYS program.
The research results showed that the relationship of kinship based on morphological characterization has the value of genetic similarity coefficient of 0.41 to 1.00 and obtained goodness of fit value amounted 0.84% (good), the value of genetic similarity coefficient on the molecular characterization was obtained between 0.76 to 1.00, and obtained goodness of fit amounted 0.91% (very good). The correlation value between morphological and molecular character of ISSR was obtained r value of -0.00840 P= < 0.321 (very weak). The results of C. reticulata accession groupings based on morphological characters were inconsistent that were obtained results from DNA band pattern analysis, so genetic similarities from morphological data cannot be used to predict the genetic similarities.
