Abstract
INDONESIA:
Adas (Foeniculum vulgare Mill.) merupakan tanaman penghasil minyak atsiri. Kandungan Minyak atsiri tanaman adas merupakan senyawa aktif bahan dasar pembuatan obat dan bahan baku industri minyak telon. Minyak atsiri dapat diperoleh melalui kultur kalus dengan penambahan zat pengatur tumbuh dan PEG 6000 yang dapat meningkatkan kandungan metabolit sekunder serta menjadi acuan konsentrasi ZPT dan PEG yang paling optimal dalam upaya meningkatkan kandungan metabolit sekunder. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian PEG 6000 terhadap kuantitas, kualitas dan kandungan metabolit sekunder pada kalus tanaman adas secara in vitro
Penelitian ini menggunakan metode deskriptif kualitatif dengan 6 perlakuan dan 5 ulangan. Perlakuan yang digunakan yaitu perbedaan pemberian konsentrasi PEG (Polyethylen Glycol) 6000 sebesar 0%, 5%, 10%, 15%, 20% dan 25%. Data pengamatan berupa data kualitatif dan kuantitatif. Data dianalisis dengan Uji ANAVA One Way α = 5%. Apabila terdapat perbedaan signifikan maka dilanjutkan uji Duncan Multiple Range Test (DMRT) dengan taraf signifikan 5%. Pengujian kandungan senyawa metabolit sekunder dilakukan menggunakan analisis Gc-Ms.
Hasil dari penelitian menunjukkan adanya pengaruh pemberian PEG 6000 terhadap kualitas, kuantitas dan kandungan metabolit sekunder kalus tanaman adas. Perlakuan PEG 6000 konsentrasi 15% menginduksi kalus pada 6hst, persentase kalus sebesar 97,5% dan berat kalus sebesar 0,32 gram. Pada perlakuan PEG terdeteksi keberadaan senyawa menthol sebesar 1,543%, anethole 1,546% dan estragol 1,746%. Pemberian PEG 6000 konsentrasi 15% dapat meningkatkan kandungan senyawa menthol sebesar 14x, kandungan senyawa anethole sebesar 4,62x dan kandungan senyawa estragol sebesar 5,35x.

Adas (Foeniculumvulgare Mill) is a plant that can produce oessential oil. Substance of essential oil in Adas is an active matter and basic material to make medicine and material of Oil for medicine. essential oil can be obtained through callus Culture with giving of growth regulator matter and PEG 6000 that can increasing substance of secondary metabolit and can be reference of ZPT consentration and PEG that so optimal on increasing substance of secondary metabolit. Purpose of this research is to know effect of giving PEG 6000 to quantity, quality and substance of secondary metabolit in adas’skalus in vitro
This research using descriptive qualitative method with 6 treatment and 5 reitration . The treatment is giving a different concentration of PEG 6000, 0%, 5%, 10%, 15%, 20% and 25%. Observation data is a qualitative data and quantitative. Analyzing data with testing the ANAVA One Way α = 5%. If there is a significant different so continued with Duncan Multiple Range Test (DMRT) with a 5% significant level. A testing of substance secondary metabolit using GC-MS analysis
The result of this research is showing that there is an effect of giving PEG 6000 to quality, quantity, and substance of secondary metabolit of fennel callus. PEG 6000 on 15% concentration inducted the callus on 6hst, callus percentage 97,5% and weight of callus is 0.32 gram. On PEG detected compound of menthol compound on 1,543%, anethole 1,546% and estragol 1.746%. giving the PEG 6000 on 15% concentration can increasing substance of mentol compound as 14-times, substance of anethole compound as 4,62-times and substance of estragol 5,35-times.
