Abstract
ABSTRAK:
Labisia “Turtle Back” dan Labisia “Black Heart” merupakan tanaman ekonomi tinggi dengan kemampuan perbanyakan alami yang terbatas, sehingga kultur in vitro diperlukan untuk mendukung propagasi secara massal. Penelitian ini bertujuan untuk mengevaluasi pengaruh 2,4-D dan picloram terhadap proliferasi kalus, tipe kalus, serta perubahan profil metabolit pada kalus yang mengalami pencoklatan. Eksplan kalus dikultur pada media MS yang mengandung BAP 0,5 mg L⁻¹ dan TDZ 0,5 mg L⁻¹ yang dikombinasikan dengan 2,4-D atau picloram pada konsentrasi 0 mg L⁻¹, 0,25 mg L⁻¹, 0,5 mg L⁻¹, dan 1 mg L⁻¹. Parameter yang diamati meliputi pertumbuhan kalus, morfologi, dan kandungan metabolit sekunder, yang dianalisis menggunakan GC–MS (Gas Chromatography–Mass Spectrometry). Respons pertumbuhan kalus menunjukkan bahwa dipengaruhi oleh genotipe dan komposisi media. Perlakuan terbaik diperoleh pada media kontrol tanpa penambahan auksin, dengan peningkatan berat segar tertinggi pada Labisia “Black Heart” sebesar 1,42 g dan laju proliferasi 3,27 ± 0,16, sedangkan Labisia “Turtle Back” mencapai laju proliferasi 2,22 ± 0,15. Penambahan 2,4-D maupun picloram cenderung menurunkan proliferasi kalus. Kalus embriogenik dicirikan bertekstur kompak dan berwarna cerah, sedangkan kalus non-embriogenik bersifat friable. Analisis GC MS menunjukkan bahwa kalus Labisia spp. didominasi oleh metabolit asam lemak. Pencoklatan kalus menyebabkan penurunan senyawa terpenoid dan hilangnya ubiquinon terpenoid seperti humulene, (+)-caryophyllene dan (-)-alpha-tocopherol.
ABSTRACT:
Labisia “Turtle Back” and Labisia “Black Heart” are ornamental plants with high economic value but limited natural propagation capacity; therefore, in vitro culture is required to support mass propagation. This study aimed to evaluate the effects of 2,4-D and picloram on callus proliferation, callus type, and changes in metabolite profiles of browned callus. Callus explants were cultured on Murashige and Skoog (MS) medium supplemented with 0.5 mg L⁻¹ BAP and 0.5 mg L⁻¹ TDZ, combined with 2,4-D or picloram at concentrations of 0 mg L⁻¹, 0.25 mg L⁻¹, 0.5 mg L⁻¹, and 1 mg L⁻¹. Parameters observed included callus growth, morphology, and secondary metabolite content using Gas Chromatography–Mass Spectrometry (GC–MS). The callus growth response was influenced by genotype and medium composition. The best treatment was obtained on control medium without auxin supplementation, with the highest fresh weight increase recorded in Labisia “Black Heart” (1.42 g) and a multiplication rate of 3.27 ± 0.16, while Labisia “Turtle Back” reached a multiplication rate of 2.22 ± 0.15. The addition of 2,4-D or picloram tended to reduce callus proliferation. Embryogenic callus was characterized by a compact texture and bright color, whereas non-embryogenic callus was friable. GC–MS analysis revealed that Labisia spp. callus was dominated by fatty acid metabolites.
