Abstract
ABSTRAK
Sirsak (Annona muricata L.) merupakan salah satu tanaman obat tradisional yang dapat dimanfaatkan sebagai obat berbagai macam antitumor. Daun sirsak mengandung bahan aktif acetogenins yang memiliki sifat sitotoksik terhadap sel tumor termasuk tumor kulit. Bahan aktif tersebut mampu mengendalikan mitokondria (organ sel penghasil energi) yang dibutuhkan sel kanker untuk berkembang. Penelitian ini bertujuan untuk mengetahui aktivitas antitumor ekstrak etanol daun sirsak (Annona muricata L.) pada kulit mencit (Mus musculus) yang diinduksi 7,12-Dimethilbenz(α)Antrasen (DMBA) secara in vivo.
Penelitian ini bersifat eksperimental menggunakan Rancangan Acak Lengkap (RAL) dengan 4 perlakuan 6 kali ulangan. Beberapa perlakuan diantaranya adalah mencit yang diinduksi DMBA 25μg/100μl aseton, diikuti dengan pemberian ekstrak etanol daun sirsak dosis 0 mg/kg BB (K+), (PI) 30 mg/kg BB, (PII) 60 mg/kg BB, dan (PIII) 90 mg/kg BB. Hewan yang digunakan mencit jantan galur Balb/c berumur 4-6 minggu dan berat 15-20 gram. Parameter yang diamati adalah preparat histopatologi jaringan kulit yang terkena tumor ditandai dengan adanya kerusakan pertumbuhan sel (displasia) dan ketebalan lapisan kulit, kejadian tumor yang meliputi jumlah mencit yang muncul nodul, volume nodul, luas permukaan luka, kerontokan bulu serta berat badan mencit. Data ketebalan lapisan kulit yang diperoleh dianalisis dengan ANOVA satu arah, apabila terdapat perbedaan sangat nyata maka dilanjutkan dengan uji duncan α 1%.
Aktivitas antitumor ekstrak etanol daun sirsak pada kulit mencit yang diinduksi DMBA dapat mempengaruhi menurunkan ketebalan lapisan kulit, jumlah mencit yang muncul nodul, mengurangi volume nodul luas permukaan luka dan kerontokan bulu serta perubahan berat badan. Dosis ekstrak etanol daun sirsak yang efektif pada penurunan ketebalan lapisan epidermis kulit mencit adalah 30mg/KgBB.
ABSTRACT
Soursop (Annona muricata L.) is one of the traditional medicinal plants that can be used as a wide range of antitumor drugs. Leaves soursop containing active ingredients acetogenins which have cytotoxic properties against tumor cells including skin tumors. The active ingredient is able to control mitochondria (energy-producing organ cells) required for cancer cells to grow. This study aims to determine the antitumor activity of the ethanol extract of leaves of soursop (Annona muricata L.) in the incidence of skin cancer mice (Mus musculus) induced 7.12-Dimethilbenz (α) Antrasen (DMBA) in vivo.
This is an experimental study using a completely randomized design (CRD) with 4 treatments 6 replications. Some treatments such as mice induced DMBA 25μg/100μl acetone, followed by ethanol extract of leaves of the soursop dose 0 mg / kg BB (K +), (PI) 30 mg / kg BB, (PII) 60 mg / kg BB, and (PIII) 90 mg / kg BB. Animals used male mice strain Balb / c aged 4-6 weeks and weighing 15-20 grams. Parameters measured were histopathological preparations exposed skin tissue tumor characterized by a defect in cell growth (dysplasia) and the thickness of the skin layer, the incidence of tumors, including the number of mice that appear nodules, nodule volume, surface area sores, hair loss and weight mice. Skin layer thickness data were analyzed by one-way ANOVA, where there is a very real difference then followed by Duncan test α 1%.
Antitumor activity of ethanol extract of leaves of the soursop in mouse skin induced DMBA can affect the lower skin layer thickness, the number of mice that appear nodules, reducing nodule volume and surface area sores hair loss and changes in body weight. Dose of ethanol extract of leaves of the soursop is effective at decreasing the thickness of the epidermal layer of the skin of mice was
30mg/KgBB.
