Abstract
INDONESIA:
Daun sirsak (Annona Muricata Linn.) diketahui memiliki aktivitas antikanker secara in-vitro. Pada penelitian ini dilakukan fraksinasi dan isolasi senyawa asetogenin dari daun sirsak yang diembankan pada zeolit NaX. Hasil pengembanan diuji aktivitas antikanker terhadap sel kanker payudara T47D.
Ekstraksi maserasi daun sirsak menggunakan pelarut etanol 95%. Fraksinasi (ekstraksi cair-cair) menggunakan campuran diklorometana: air dilanjutkan dengan n-heksana: metanol. Pemisahan dengan KLTA untuk mengetahui eluen terbaik dan pemisahan dengan KLTP untuk mendapatkan isolat menggunakan eluen metanol: diklorometana (1:19). Ekstrak hasil fraksinasi dan isolat hasil KLT diembankan pada zeolit NaX menggunakan metode impregnasi kering dengan perbandingan berat 5:10. Keberhasilan pengembanan dianalisis menggunakan spektrofotometer FTIR. Metode uji aktivitas antikanker yang digunakan adalah metode MTT (Microculture tetrazolium).
Hasil analisis spektrofotometer FTIR menunjukkan bahwa sampel ekstrak hasil fraksinasi: zeolit, muncul puncak serapan baru pada bilangan gelombang 2909,881 cm-1 untuk gugus Csp3-H (stretch) asimetri dan 2851,383 cm-1 untuk - Csp3-H (stretch) simetri. Sampel isolat hasil KLT: zeolit, puncak serapan baru muncul pada bilangan gelombang 2935,178 cm-1 untuk gugus Csp3-H (stretch) asimetri dan 2851,383 cm-1 untuk Csp3-H (stretch) simetri. Munculnya serapan baru menunjukkan bahwa senyawa telah teremban dalam zeolit. Nilai IC50 yang diperoleh dari sampel pengembanan ekstrak hasil fraksinasi dan isolat hasil KLT pada zeolit NaX yaitu 125,683 g/mL dan 103.514,217 g/mL.
ENGLISH:
The soursop leaf (Annona muricata Linn.) is known to have anticancer activity in vitro. Fractionation and isolation of acetogenin compound from soursop leaf that impregnated to zeolite NaX were carried out in this study. The result were tested for anticancer activity on T47D breast cancer cells.
Maceration of soursop leaves using 95% ethanol. Fractionation (liquid-liquid extraction) using a mixture of dichloromethane: water followed by n-hexane: methanol. Separation with analitycal-TLC to find out the best eluent and separation with preparative-TLC to get isolates using eluent methanol: dichloromethane (1:19). Extract from fractination and isolate from TLC were carried out on NaX zeolite using the dry impregnation method with a ratio of 5:10. The result of impregnation was analyzed using FTIR spectrophotometer. The anticancer activity test using MTT (Microculture tetrazolium) method.
The FTIR spectrophotometer results showed that extract from fractination: zeolite, new absorption peaks appeared at wavenumber 2909.881 cm-1 for Csp3-H (stretch) asymmetry and 2851,383 cm-1 for Csp3-H (stretch) symmetry. Isolate from TLC: zeolite sample, the new absorption peak appeared at wavenumber 2935,178 cm-1 for the Csp3-H (stretch) asymmetry group and 2851,383 cm-1 for - Csp3-H (stretch) symmetry. The appearance of new absorption shows that the compound has been impregnated in zeolite. The IC50 value of extract from fractination and isolates from TLC sample that has been impregnated in NaX zeolite was 125,683 g/mL and 103,514,217 g/mL.
