Abstract

INDONESIA:

Semakin berkembangnya kesadaran masyarakat tentang kesehatan, kini pangan dituntut untuk mempunyai nutrisi yang tinggi dan harus mempunyai nilai yang bermanfaat bagi tubuh. Salah satu pangan yang sedang dikembangkan saat ini adalah pangan probiotik. Tujuan penelitian ini adalah mengetahui potensi Lactobacillus plantarum sebagai probiotik.

Penelitian ini dilaksanakan di Laboratorium Mikrobiologi, jurusan Biologi Fakultas Sains dan Teknologi, Universitas Islam Negeri Maulana Malik Ibrahim Malang. Mulai bulan September sampai Desember 2010. Metode penelitian yang digunakan adalah metode eksperimen secara deskriptif yang bertujuan untuk memberikan informasi tentang potensi probiotik dari Lactobacillus plantarum dengan pengujian secara in vitro yaitu kajian ketahanan terhadap asam, garam empedu 0,3% (b/v) dan penghambatan terhadap bakteri patogen, yaitu Staphylococcus aureus, Escherechia coli dan Salmonella thypi.

Data yang diperoleh dari penelitian ini dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa seiring dengan kenaikan pH maka jumlah Lactobacillus plantarum yang hidup semakin meningkat. Rata-rata jumlah Lactobacillus plantarum yang tumbuh pada pH 2 adalah 2,7 . 107 cfu/ml, pada pH 3 sebanyak 3. 107 cfu/ml dan pada pH 4 sebanyak 4,9 . 107 cfu/ml, sedangkan pada kontrol sebanyak 5,3. 108 cfu/ml. Hasil uji ketahanan Lactobacillus plantarum terhadap garam empedu 0,3% (b/v) menunjukkan bahwa Lactobacillus plantarum mempunyai ketahanan terhadap garam empedu 0,3% (b/v). Rata-rata jumlah Lactobacillus plantarum yang tumbuh adalah 5,5. 109 cfu/ml, sedangkan pada kontrol sebanyak 2,9. 1010 cfu/ml. Hasil uji penghambatan Lactobacillus plantarum terhadap bakteri patogen menunjukkan bahwa pengujian terhadap Staphylococcus aureus rata-rata diameter penghambatan sekitar 6,3 mm, terhadap Salmonella typhi menunjukkan diameter penghambatan sekitar 4 mm dan terhadap Escherecia coli menunjukkan rata-rata diameter penghambatan sekitar 3,3 mm.

Progressively expanding awareness of the society about health, nowdays food claimed to have high nutrition and have to have worthwhile value to body. One kind of developing food today is probiotics. Aim of this research is to know potential of Lactobacillus plantarum as proboitic.

This research was conducted at the Laboratory of Microbiology, Department of Biology, Faculty of Science and Technology, Islamic State University Maulana Malik Ibrahim of Malang. From September to December 2010. The research method used is descriptive method of experiment which aims to provide information about the potential of probiotic Lactobacillus plantarum with the in vitro testing of the study resistance to acids, bile salts 0.3% (w/v) and inhibition of pathogenic bacteria, namely Staphylococcus aureus, Escherechia coli and Salmonella thypi.

Data obtained from this study were analyzed descriptively. The results showed that as pH increased the number of Lactobacillus plantarum living increases. The average number of Lactobacillus plantarum grown at pH 2 is 2,7. 107 cfu/ml, at pH 3 of 3. 107 cfu/ml and at pH 4 as much as 4,9. 107 cfu/ml, while in control as much as 5,3. 108 cfu/ml. Lactobacillus plantarum resistance test result of 0.3% bile salt (w/v) showed that Lactobacillus plantarum has the resistance to bile salts 0.3% (w/v). The average number of Lactobacillus plantarum growing was 5,5. 109 cfu/ml, while in control as much as 2,9. 1010 cfu/ml. The results inhibition of Lactobacillus plantarum against pathogenic bacteria indicates that the average inhibition diameter of Staphylococcus aureus about 6,3 mm, the average inhibition diameter of Salmonella typhi about 4 mm and the average inhibition diameter of Escherecia coli about 3,3 mm.
