Abstract
INDONESIA:
Natrium diklofenak merupakan obat golongan Anti Inflamatory Non Steroid (AINS) yang berkhasiat sebagai analgesik, antipiretik dan antiinflamasi. Apabila dikonsumsi secara oral, natrium dikofenak mengalami First Past Effect (FPE) sekitar 50% dan berefek samping pada sistem gastrointestinal. Natrium diklofenak dibentuk sediaan topikal untuk menghindari hal tersebut. Sediaan topikal yang paling baik untuk natrium diklofenak yaitu gel. Gelling agent dan enhancer merupakan bahan tambahan yang berperan penting dalam sediaan gel. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh konsentrasi gelling agent carbopol 940, konsentrasi enhancer asam oleat dan interaksi keduanya dalam menentukan respon viskositas dan fluks pelepasan natrium diklofenak dalam basis gel serta untuk menemukan komposisi optimum carbopol 940 dan asam oleat yang dapat memberikan respon viskoisitas antara 20 dPa.s - 200 dPa.s dan respon fluks pelepasan antara 50 μg/cm2.menit1/2 – 150 μg/cm2.menit1/2.
Penelitian ini merupakan rancangan eksperimental laboratorik menggunakan desain faktorial dua faktor dan dua level yaitu level rendah dan level tinggi. Faktor carbopol 940 berlevel rendah 0,8% dan berlevel tinggi 1,3%, sedangkan faktor asam oleat berlevel rendah 5% dan berlevel tinggi 20%. Analisis pengaruh (efek) kedua faktor terhadap respon serta penentuan formula optimum dianalisis menggunakan software Design Expert 10. Uji Anova dilakukan untuk mengetahui signifikansi efek faktor yang didapat terhadap respon.
Hasil penelitian menunjukkan bahwa carbopol 940 dan asam oleat berpengaruh signifikan dalam menentukan respon viskositas dan fluks pelepasan. Penambahan carbopol 940 dan asam oleat akan meningkatkan respon viskositas dan penambahan carbopol 940 dan asam oleat akan menurunkan respon fluks pelepasan. Terdapat 100 komposisi optimum kombinasi carbopol 940 antara 0,8% sampai 1,194% dan asam oleat antara 5% sampai 19,95% yang dapat digunakan untuk memperoleh sediaan gel natrium diklofenak dengan respon viskositas antara 76,042 dPa.s –199,570 dPa.s dan respon fluks pelepasan antara 101,000 µg/cm2.menit1/2–124,250 µg/cm2.menit1/2

Diclofenac sodium is categorized as Anti Inflammatory Non-Steroid (AINS) and used as analgesic, antipyretic and antiinflammation. When it is consumed orally, diclofenac sodium undergoes First Past Effect (FPE) around 50% and has side effect on gastrointestinal track. In order to prevent it, diclofenac sodium then formed into topical preparation. The best topical preparation for diclofenac natrium is gel. Gelling agent and enhancer is the additional material which are fundamental for the gel preparation. The purpose of this research is to find out the influence of Carbopol 940 gelling agent concentration, oleic acid concentration enhancer and the interaction between them in deciding the respons of viscosity and flux of diclofenac sodium release in gel-based, also to find out the optimum composition of Carbopol 940 and oleic acid which respond to viscosity between 20dPa.s – 200dPa.s and response of flux release between 50 μg/cm2.minutes1/2 – 150 μg/cm2.minutes1/2.
This research is considered as laboratory experimental plan using factorial design with two factors and two levels, low and high level. The low level of Carbopol 940 factor is 0.8% and the high level is 1.3%, while low level of oleic acid factor is 5% and the high level is 20%. The effect analysis of bot factors toward the response and formula optimum determination was analyzed using Design Expert 10 software. Anova test was conducted to find out the significance of factor effect obtained towards the response.
The result of this research shows that Carbopol 940 and oleic acid have a significance influence in determining the viscosity and flux release response. Addition of Carbopol 940 and oleic acid will increase the viscosity response and addition of Carbopol 940 and oleic acid will decrease flux release response. There are 100 optimum combinations composition of Carbopol 940 between 0.8% up to 1.194% and oleic acid between 5% up to 19.95% which can be used to obtain diclofenac sodium gel preparation with viscosity response between 76.042 dPa.s – 119.570 dPa.s and flux release response between 10.1000 µg/cm2.minutes/2–124,250 µg/cm2.minutes1/2
