Abstract
INDONESIA:
Benalu mangga (Dendrophthoe pentandra) merupakan tanaman semiparasit terhadap inangnya yang berpotensi sebagai tumbuhan obat, sehingga diperlukan metabolite profiling untuk mengetahui senyawa-senyawa yang terdapat dalam daun benalu mangga. Tujuan penelitian ini adalah untuk mengetahui perbedaan profil metabolit daun benalu mangga yang diperoleh dari daerah Kediri, Jawa Timur; Pekalongan, Jawa Tengah; Gunung Batin Baru, Lampung dan Tanjung, Selor Hilir, Kalimantan Utara.
Metode ekstraksi dengan bantuan ultrasonik dengan pelarut etanol 96% perbandingan 1:10 [b/v] digunakan untuk memperoleh ekstrak kental daun benalu mangga. Keempat ekstrak dari daerah yang berbeda dianalisis menggunakan UPLC-QToF-MS/MS fase gerak campuran air/asam format 99,9/0,1 [v/v] dan asetonitril/asam format 99,9/0,1 [v/v] dengan sistem elusi gradien dan fase diam C18. Hasil dari analisis tersebut dilanjutkan dengan analisis kemometrik menggunakan PCA menggunakan perangkat lunak Minitab versi 17.
Terdapat perbedaan metabolit antar keempat daerah perolehan ekstrak. Pengelompokkan berdasarkan metabolit yang hampir serupa terjadi pada daerah Kediri, Jawa Timur dengan daerah Tanjung Selor Hilir, Kalimantan Utara. Diduga senyawa methyldioctylamine; scortechinone F; 3-Cyclohexyl-N-(ethoxycarbonyl)-L-alanyl-N-[(4S,5E,7R)-7-carbamoyl-9-methyl-5-decen-4-yl]-L-lysinamide; dan Pheophorbide A merupakan senyawa penciri berdasarkan asal geografisnya.
ENGLISH:
Benalu mangga (Dendrophthoe pentandra) is a semi-parasitic plant which is potential as a medicinal plant, and it needs metabolite profiling to find out the compounds contained in that leaves. The purpose of this research is to find out the difference of metabolite profile of benalu mangga leaves obtained from Kediri, East Java; Pekalongan, Central Java; Gunung Batin Baru, Lampung and Tanjung, Selor Hilir, North Kalimantan.
Ultrasonic assisted extraction with 96% ethanol as solvent, ratio of 1:10 [b/v] was used to obtain concentrated extract of benalu mangga leaves. The four extracts from different regions was analyzed using UPLC-QToF-MS/MS. UPLC system were performed by C18 as stationary phase and gradient elucidation water/formic acid 99,9/0,1 [v/v]-acetonitrile/formic acid 99,9/0,1 [v/v]. The results of that analysis were analyzed by chemometric analysis using PCA Minitab software version 17.
There are differences on metabolite obtained from the four sampling areas. Grouping based on the metabolite similiarity is occurred in Kediri, East Java with Tanjung Selor Hilir, North Kalimantan. It is suspected that methyldioctylamine; scortechinone F; 3-Cyclohexyl-N-(ethoxycarbonyl)-L-alanyl-N-[(4S,5E,7R)-7-carbamoyl-9-methyl-5-decen-4-yl]-L-lysinamide; and Pheophorbide A are biomarker compound based on its geographic origin.
