Abstract

INDONESIA:

Infeksi Salmonella typhimurium menjadi masalah kesehatan global karena kemampuan bakteri ini membentuk biofilm yang meningkatkan resistensi terhadap antibiotik. Apel manalagi (Malus sylvestris) mengandung senyawa bioaktif seperti flavonoid, alkaloid, tanin, saponin, dan steroid yang berpotensi sebagai antibiofilm. Penelitian ini bertujuan mengetahui aktivitas antibiofilm dekok buah apel manalagi terhadap Salmonella typhimurium melalui uji pencegahan perlekatan, penghambatan pertumbuhan, dan penghancuran biofilm. Penelitian eksperimental murni (true experimental) dengan rancangan post test only control group design ini menggunakan sampel dekok buah apel manalagi konsentrasi 50%, 25%, 12,5%, dan 6,25% serta bakteri Salmonella typhimurium ATCC-14028. Pengujian aktivitas antibiofilm menggunakan metode microtiter plate biofilm assay dengan pewarnaan crystal violet dan pengukuran optical density (OD) pada panjang gelombang 595 nm. Analisis data menggunakan uji One-Way ANOVA dilanjutkan Post Hoc, uji korelasi Pearson, dan uji regresi linier sederhana. Hasil penelitian menunjukkan bahwa dekok buah apel manalagi pada konsentrasi 6,25% memiliki efektivitas pencegahan perlekatan biofilm (OD=0,153; 84,53%) dan penghambatan pertumbuhan biofilm (OD=0,212; 83%) yang sebanding dengan kontrol positif (p>0,05). Pada uji penghancuran biofilm, semua konsentrasi (6,25%-50%) efektif dibandingkan kontrol negatif (p<0,05), namun tidak terdapat perbedaan signifikan antar konsentrasi. Uji fitokimia mengonfirmasi kandungan flavonoid, alkaloid, tanin, saponin, dan steroid dalam dekok. Simpulannya, dekok buah apel manalagi memiliki aktivitas antibiofilm terhadap Salmonella typhimurium dengan efektivitas optimal pada konsentrasi 6,25% untuk pencegahan perlekatan dan penghambatan pertumbuhan biofilm.

Salmonella typhimurium infection is a global health problem due to the bacterium's ability to form biofilms that increase antibiotic resistance. Manalagi apple (Malus sylvestris) contains bioactive compounds such as flavonoids, alkaloids, tannins, saponins, and steroids that have potential as antibiofilm agents. This study aimed to determine the antibiofilm activity of Manalagi apple decoction against Salmonella typhimurium through biofilm prevention, inhibition, and eradication assays. This true experimental study with a post test only control group design used Manalagi apple decoction at concentrations of 50%, 25%, 12.5%, and 6.25% and Salmonella typhimurium ATCC-14028 bacteria. Antibiofilm activity was tested using the microtiter plate biofilm assay with crystal violet staining and optical density (OD) measurement at 595 nm wavelength. Data were analyzed using One-Way ANOVA followed by Post Hoc test, Pearson correlation test, and simple linear regression. The results showed that Manalagi apple decoction at 6.25% concentration had biofilm prevention (OD=0.153; 84.53%) and biofilm inhibition (OD=0.212; 83%) effectiveness comparable to the positive control (p>0.05). In the biofilm eradication assay, all concentrations (6.25%-50%) were effective compared to the negative control (p<0.05), but no significant differences were found between concentrations. Phytochemical analysis confirmed the presence of flavonoids, alkaloids, tannins, saponins, and steroids in the decoction. In conclusion, Manalagi apple decoction exhibits antibiofilm activity against Salmonella typhimurium with optimal effectiveness at 6.25% concentration for biofilm prevention and inhibition.
