Abstract
ABSTRAK:
Acrylic acid merupakan material yang dalam bentuk polimer disebut poly acrylic acid banyak dimanfaatkan sebagai material drug delivery system (DDS) karena sifat bioadhesif dan kemampuannya membentuk jaringan crosslink akibat paparan radiasi gamma. Cross-link pada poly acrylic acid terjadi akibat radiolisis gugus karboksil (–COOH). Pemahaman mengenai mekanisme radiolisis gugus karboksil (–COOH) pada acrylic acid menjadi sangat penting untuk menentukan kondisi iradiasi yang optimal dalam pengembangan material DDS berbasis polimer. Penelitian ini bertujuan untuk menganalisis model geometri monomer, dimer, dan trimer acrylic acid yang memiliki energi paling rendah, menganalisis perubahan kerapatan elektron pada ikatan C–C dan O–H akibat ionisasi, serta mengidentifikasi mekanisme radiolisis gugus karboksil yang terjadi pada acrylic acid. Simulasi dilakukan secara komputasi menggunakan metode Density Functional Theory (DFT) dengan fungsional CAM-B3LYP/6-311++G** yang dikombinasikan dengan Ab Initio Molecular Dynamics (AIMD) pada perangkat lunak ORCA 6.0.0, terhadap model acrylic acid dalam berbagai kondisi ionisasi. Hasil optimasi geometri menunjukkan panjang ikatan C–C sebesar 1,51 Å dan O–H sebesar 0,96 Å pada ketiga model, konsisten dengan nilai perhitungan eksperimen. Analisis distribusi elektron menunjukkan bahwa penurunan kerapatan elektron akibat ionisasi berpusat pada gugus –COOH dan ikatan C–C mengalami penurunan kerapatan elektron seiring meningkatnya tingkat ionisasi, sementara ikatan O–H tidak menunjukkan penurunan yang signifikan. Hasil keseluruhan ini menunjukkan bahwa mekanisme radiolisis pada acrylic acid yaitu ketika acrylic acid dikenai radiasi gamma dan mengalami ionisasi, dan akan mengalami penurunan kerapatan elektron yang menyebabkan disosiasi ikatan C–C, tapi tidak dengan ikatan O–H, hasil ini sedikit berbeda dengan temuan mekanisme radiolisis sebelumnya, dikarenakan tidak adanya interface antara molekul acrylic acid dengan molekul lain dalam simulasi ini, sehingga ikatan O–H konsisten tidak mengalami pemutusan. Temuan ini mengindikasikan bahwa radiasi gamma secara selektif memutus ikatan C–C untuk membentuk crosslink sambil mempertahankan keutuhan gugus –COOH yang bertanggung jawab terhadap sifat bioadhesif acrylic acid sebagai material DDS.
ABSTRACT:
Acrylic acid, which in its polymeric form is known as poly(acrylic acid), is widely used as a drug delivery system (DDS) material due to its bioadhesive properties and its ability to form cross-links upon exposure to gamma radiation. Cross-linking in poly(acrylic acid) is associated with the radiolysis of the carboxyl group (–COOH). Therefore, understanding the mechanism of radiolysis of the carboxyl group in acrylic acid is essential for determining the optimal irradiation conditions for the development of polymer-based DDS materials. This study aims to analyze the lowest-energy geometric structures of acrylic acid monomers, dimers, and trimers, investigate changes in electron density at the C–C and O–H bonds induced by ionization, and identify the radiolysis mechanism of the carboxyl group in acrylic acid. Computational simulations were performed using Density Functional Theory (DFT) with the CAM-B3LYP/6-311++G** method combined with Ab Initio Molecular Dynamics (AIMD) as implemented in ORCA 6.0.0. Acrylic acid models were examined under various ionization conditions. Geometric optimization results showed C–C bond lengths of 1.51 Å and O–H bond lengths of 0.96 Å for all three models, in good agreement with experimental values. Electron density difference analysis revealed that the reduction in electron density caused by ionization is primarily localized within the –COOH group. Furthermore, the C–C bond exhibited a progressive decrease in electron density with increasing ionization level, whereas the O–H bond showed no significant reduction. These findings suggest that the C–C bond is susceptible to dissociation upon ionization, while the O–H bond remains relatively stable. AIMD simulations further demonstrated that radiolysis in acrylic acid proceeds through a direct ionization pathway that initiates C–C bond cleavage (chain scission) as the first step of the radiolysis process. In contrast, the O–H bond did not undergo cleavage under any of the simulated ionization conditions. The AIMD results are consistent with the electron density analysis, confirming that ionization promotes C–C bond dissociation but does not induce O–H bond cleavage. Overall, the results indicate that when acrylic acid is exposed to gamma radiation and undergoes ionization, the resulting decrease in electron density leads to selective C–C bond dissociation while preserving the O–H bond. This radiolysis mechanism differs slightly from previous findings, as the absence of an interface prevents O–H bond cleavage. These findings suggest that gamma radiation selectively breaks C–C bonds to initiate cross-link formation while maintaining the integrity of the –COOH groups responsible for the bioadhesive properties of acrylic acid as a DDS material

حض األكريليك هو مادة تُعرف يف شكل بوليمر ابسم بويل أكريليك أسيد، وتُستخدم كثيًا كمادة يف نظام توصيل الدواء)DDS(بسبب خصائصها الالصقة احليوية وقدرهتا على تكوين شبكات متشابكة نتيجة كسيلالتعرض إلشعاع غاما. حيدث الرتابط املتقاطع يف بويل أكريليك أسيد نتيجة لتفكك جمموعات الكربو)- COOH(كسيلابإلشعاع. فهم آلية تفكك جمموعات الكربو)-COOH(يف محض األكريليك مهم جدًا لتحديد ظروف اإلشعاع املثلى يف تطوير موادDDSالقائمة على البوليمر. هتدف هذه الدراسة إىل حتليل منوذج اهلندسة ملونومرات، ثنائيات، وثالثيات محض األكريليك اليت متتلك أقل طاقة، وحتليل التغيات يف كثافة اإللكرتوانت يف الروابطC–CوO–Hكسيل احلاصلة يف محض األكريليككذلك حتديد آلية تفكك جمموعات الكربوبسبب التأين، و .مت إجراء احملاكاة حسابيًا ابستخدام طريقة نظرية الكثافة الدالية)DFT(مع الدالة الوظيفيةCAM-B3LYP/6- 311++G**املدجمة مع الديناميكيا اجلزيئية من املنهج الغزوي)AIMD(على برانمجORCA6.0.0، على منوذج محض األكريليك يف ظروف أتين خمتلفة. أظهرت نتائج حتسني اهلندسة طول الرابطةC–Cمبقدار1.51Åوالرابطة O–Hمبقدار0.96Åيف النماذج الثالثة، وهو ما يتوافق مع قيم احلساابت التجريبية. أظهر حتليل فرق كثافة كز حول اجملموعةكثافة اإللكرتون بسبب التأين يتمر اإللكرتون أن اخنفاض–COOHوأن الرابطةC–Cتتعرض كثافة اإللكرتون مع ازدايد مستوى التأين، بينما مل تظهر الرابطة الخنفاضO–Hكبيًا. تُظهر هذه النتائج اخنفاضًا أن الرابطةC–Cأكثر عرضة للتفكك بسبب التأين، ولكن ليس الرابطةO–H.تُظهر نتائج حماكاةAIMDأن حتليل الروابط بني الكربون-الكربون حيدث نتيجة التأيني، كما تنبأت نتائج كثافة اإللكرتوانت، كما أن نتائج AIMDتشي أيضًا إىل أن روابط الكربون-الكربون تتعرض لالنفصال بسبب التأيني، ولكن ليس للروابطO-H. تشي هذه النتائج بشكل عام إىل أن آلية حتليل اإلشعاع يف محض األكريليك هي أنه عند تعرض محض األكريليك كثافة اإللكرتوانت مما يؤدي إىل انفصال روابط لإلشعاع املؤين، تتناقصC-C، ولكن ال حيدث ذلك مع روابط O-H.هذه النتائج ختتلف قليال ً عن االكتشافات السابقة آللية حتليل اإلشعاع، بسبب عدم وجود واجهة، لذلك ال تتعرض روابطO-Hلالنكسار. وتشي هذه النتائج إىل أن اإلشعاع الغاما يكسر بشكل انتقائي روابطC-C لتشكيل وصالت متقاطعة مع احلفاظ على سالمة جمموعة–COOHاملسؤولة عن اخلصائص الالصقة البيولوجية كمادة حلمض األكريليكD
