Abstract

INDONESIA

Bakteri patogen secara global memiliki prevalensi infeksi yang cukup tinggi terhadap manusia, yaitu Staphylococcus aureus (30%) dan Escherichia coli (16,5%). Pengobatan infeksi menggunakan antibiotik secara tidak tepat dapat memicu resistensi antibiotik sehingga dibutuhkan obat yang lebih efektif. Salah satunya yaitu N-(5-(methylthio)-1,3,4-thiadiazol-2-yl)-4-(trifluoromethyl) benzamide. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri senyawa N-(5-(methylthio)-1,3,4-thiadiazol-2-yl)-4-(trifluoromethyl) benzamide terhadap Staphylococcus aureus dan Escherichia coli berdasarkan nilai Minimum Inhibitory Concentration (MIC) dan klasifikasi zona hambat. Metode uji aktivitas antibakteri menggunakan uji difusi cakram dengan media Mueller Hinton Agar. Penelitian ini memiliki 5 kelompok perlakuan, yaitu 3 kelompok uji dengan konsentrasi 125 µg/ml, 100 µg/ml, 75 µg/ml serta kelompok kontrol positif Ciprofloxacin 25 µg/ml dan kontrol negatif DMSO yang direplikasi sebanyak 5 kali. Inhibitory Concentration (MIC) sebesar 75 µg/ml dan rata-rata diameter zona
hambat sebesar 7,76 mm (75 µg/ml), 9,73 mm (100 µg/ml) dan 9,84 mm (125 µg/ml) dengan klasifikasi yang bersifat lemah. Sementara hasil uji aktivitas antibakteri senyawa uji terhadap Escherichia coli memiliki Minimum Inhibitory Concentration (MIC) sebesar 75 µg/ml dan rata-rata diameter zona hambat sebesar 7,87 mm (75 µg/ml), 8,59 mm (100 µg/ml) dan 9,65 mm (125 µg/ml) dengan klasifikasi yang bersifat lemah. Kesimpulannya adalah senyawa N-(5-(methylthio)1,3,4-thiadiazol-2-yl)-4-(trifluoromethyl)benzamide memiliki aktivitas antibakteri terhadap Staphylococcus aureus dan Escherichia coli dengan klasifikasi yang bersifat lemah

Pathogenic bacteria have a relatively high global prevalence of infection in humans, particularly Staphylococcus aureus (30%) and Escherichia coli (16.5%). Inappropriate use of antibiotics in the treatment of infections can lead to antibiotic resistance; therefore, more effective therapeutic agents are needed. One such potential compound is N-(5-(methylthio)-1,3,4-thiadiazol-2-yl)-4-(trifluoromethyl)benzamide. This study aimed to determine the antibacterial activity of N-(5-(methylthio)-1,3,4-thiadiazol-2-yl)-4-(trifluoromethyl)benzamide against Staphylococcus aureus and Escherichia coli based on Minimum Inhibitory Concentration (MIC) values and inhibition zone classification. The antibacterial activity test was conducted using the disc diffusion method on Mueller Hinton Agar. The study consisted of five treatment groups, including three test groups with concentrations of 125 µg/mL, 100 µg/mL, and 75 µg/mL, a positive control group using ciprofloxacin 25 µg/mL, and a negative control group using DMSO, with five replications for each group. The antibacterial activity test results against Staphylococcus aureus showed a Minimum Inhibitory Concentration (MIC) of 75 µg/mL and mean inhibition zone diameters of 7.76 mm (75 µg/mL), 9.73 mm (100 µg/mL), and 9.84 mm (125 µg/mL), which were classified as weak. Meanwhile, the antibacterial activity test against Escherichia coli showed a Minimum Inhibitory Concentration (MIC) of 75 µg/mL and mean inhibition zone diameters of 7.87 mm (75 µg/mL), 8.59 mm (100 µg/mL), and 9.65 mm (125 µg/mL), which were also classified as weak. In conclusion, N-(5-(methylthio)-1,3,4-thiadiazol-2-yl)-4-(trifluoromethyl)benzamide exhibits antibacterial activity against Staphylococcus aureus and Escherichia coli with a weak inhibition classification.
