Abstract
INDONESIA:
Mikroalga merupakan kelompok tumbuhan berukuran renik yang hidup di seluruh wilayah perairan, baik air tawar maupun air laut. Mikroalga mengandung banyak senyawa aktif yang dapat dimanfaatkan oleh manusia sebagai obat. Penelitian ini bertujuan untuk mengetahui tingkat toksisitas fraksi etil asetat, kloroform, petroleum eter dan n-heksana hasil hidrolisis ekstrak metanol mikroalga Chlorella sp. terhadap larva udang Artemia salina Leach serta mengetahui golongan senyawa aktif yang memiliki toksisitas tertinggi terhadap Artemia salina Leach.
Ekstraksi senyawa aktif dilakukan dengan metode maserasi menggunakan pelarut metanol p.a.. Ekstrak metanol dihidrolisis dengan larutan HCl 2 N kemudian dipartisi bertingkat dengan empat variasi pelarut yaitu n-heksana, petroleum eter, kloroform dan etil asetat. Masing-masing fraksi diuji toksisitasnya terhadap larva udang Artemia salina Leach. Data kematian larva udang dianalisis dengan analisis probit untuk mengetahui nilai LC50. Semua fraksi diidentifikasi senyawa aktifnya dengan menggunakan uji reagen. Fraksi yang memiliki toksisitas tertinggi dipisahkan senyawa aktifnya dengan menggunakan kromatografi lapis tipis.
Hasil penelitian menunjukkan bahwa ekstrak petroleum eter memiliki toksisitas tertinggi terhadap larva udang Artemia salina Leach (LC50 rendah). Nilai LC50 masing- masing ekstrak yaitu: ekstrak metanol (38,9697 ppm), fraksi etil asetat (43,3044 ppm), fraksi kloroform (32,9023 ppm), fraksi petroleum eter (32,6710 ppm), fraksi n-heksana (34,2133 ppm). Microalgae are microscopic-sized groups of plants that live around waters area, both fresh water and sea water. Microalgae contain many active compounds that can be used by humans as a drug. The objectives of this study are to know the level of toxicity ethyl acetate fraction, chloroform, petroleum ether and n-hexane hydrolysis result of methanol extracts microalgae Chlorella sp. toward larvae shrimp of Artemia salina Leach and to know the active compounds that has highest toxicity to Artemia salina Leach.
Extraction of active compounds is done by maceration method using methanol p.a.. Methanol extract hydrolyzed with HCl 2 N and then partitioned storey with four variations solvent those are n-hexane, petroleum ether, chloroform and ethyl acetate. Each fraction was tested the toxicity toward larvae shrimp of Artemia salina Leach. Mortality data of Larvae Shrimp were analyzed by probit analysis to determine LC50 values. All fractions active compounds identified by using reagent test. Fraction that has highest toxicity of active compounds separated by thin layer chromatography.
The results of this study showed that the petroleum ether fraction had the highest toxicity to larvae shrimp of Artemia salina Leach (lower LC50). LC50 values of each extract are: methanol extract (38.9697 ppm), ethyl acetate fraction (43.3044 ppm), chloroform fraction (32.9023 ppm), petroleum ether fraction (32.6710 ppm), n-hexane fraction (34.2133 ppm). Identification of the active compound group with reagent test showed that the petroleum ether fraction contained steroid compounds.
