Abstract
INDONESIA:
Al Quran surat as Sajdah ayat 27 menjelaskan bahwa Allah SWT menumbuhkan tanaman sebagai tanda kekuasaan-Nya sebagai bahan berfikir sehingga dapat dimanfaatkan, salah satunya untuk pengobatan. Pada penelitian ini telah dilakukan isolasi dan uji efektivitas antimalaria isolat alkaloid tanaman Anting-anting secara in vivo pada mencit jantan. Penelitian ini bertujuan mencari eluen terbaik dalam pemisahan ekstrak kasar alkaloid dengan metode KLT serta mengetahui aktivitas antimalaria isolat alkaloid terhadap pertumbuhan P. berghei.
Isolasi alkaloid tanaman Anting-anting dilakukan dengan maserasi menggunakan metanol dan ekstraksi cair-cair secara asam basa. Ekstrak alkaloid diuji fitokimia menggunakan reagen dan dipisahkan dengan KLT analitik untuk mencari eluen terbaik dengan variasi eluen kloroform : metanol (9,5:0,5), kloroform : metanol : amoniak (85:15:1), kloroform : metanol (1:4), etil asetat : metanol : air (6:4:2) dan kloroform : etil asetat (8:2). Hasil isolat alkaloid diuji aktivitas antimalaria in vivo terhadap mencit jantan. Data derajat parasitemia dianalisis dengan Uji One Way ANOVA dan uji Tukey.
Hasil KLT analitik menunjukkan bahwa eluen terbaik untuk KLT Preparatif adalah kloroform:metanol (9,5:0,5) (v/v) yang menghasilkan 7 noda dan berwarna jingga, coklat dan kuning kehijauan setelah disemprot reagen Dragendorf dan dideteksi di bawah lampu UV 366 nm dengan Rf antara 0,21-0,92. Berdasarkan analisis Uji One Way ANOVA dan uji Tukey diperoleh kesimpulan bahwa antara ketujuh isolat alkaloid jika dibandingkan dengan kontrol negatif tidak ada perbedaan yang signifikan. Akan tetapi, kelompok perlakuan isolat alkaloid 4 dan 6 memiliki % penghambatan parasit, berturut-turut sebesar 21,2 % (b/b) dan 11,9 % (b/b).

Al Quran surah as Sajdah Verse 27 has explained that God grows the plant as the sign of God existence to keep His creatures thinking about the benefits. One benefit of plant is for medication. This research, therefore, attempts to do the isolation and the in-vivo anti-malaria effectiveness test of Anting-Anting Plant in the Male Mouse. The objective of research is to search for the best eluent by isolating alkaloid crude extract with TLC method and to understand the anti- malaria activity of alkaloid isolate against P.berghei growth.
The isolation of Anting-Anting based alkaloid is conducted with maceration involving methanol and acid-alkali based liquid-liquid extraction. Alkaloid extract is subjected to phytochemical test using reagent and then, it is isolated with analytical TLC to produce the best eluent with the following eluent variations: chloroform : methanol (9.5:0.5), chloroform : methanol : ammonia (85:15:1), chloroform : methanol (1:4), acetate ethyl : methanol : water (6:4:2), and chloroform : acetate ethyl (8:2). The resultant of alkaloid isolate is tested for in-vivo anti-malaria activity in the male mouse. Data of parasitemia degree is analyzed with One-Way ANOVA Test and Tukey Test.
Result of Analytic TLC indicates that the best eluent for Preparative TLC is chloroform : methanol (9.5:0.5) (v/v) which produces 7 stains. Orange red, brown, and greenish yellow are colors emerging after the eluent is sprayed with Dragendorf Reagent and detected below 366 nm UV lamp with Rf between 0.21- 0.92. Result of One Way ANOVA indicates that seven alkaloid isolates have very low potentials in preventing the growth of P. Berghei parasite compared to negative control group. However, the exception is found because Group 4 and 6 of alkaloid isolate treatment have prevention against parasite for 21.2 % (w/w) and 11.9 % (w/w).
