Abstract
INDONESIA :
Shigellosis akibat infeksi Shigella flexneri merupakan masalah kesehatan global yang berhubungan dengan kerusakan mukosa usus melalui peningkatan apoptosis sel epitel. Ekstrak kayu manis (Cinnamomum verum) mengandung senyawa antibakteri, antiinflamasi, dan antioksidan yang berpotensi menekan apoptosis berlebihan pada epitel usus. Penelitian ini bertujuan mengetahui pengaruh ekstrak kayu manis terhadap apoptosis sel epitel ileocaecal junction mencit (Mus musculus L.) yang diinduksi Shigella flexneri. Penelitian eksperimental ini menggunakan rancangan post-test only control group dengan Rancangan Acak Lengkap. Mencit Balb/c betina dibagi menjadi kelompok kontrol negatif, kontrol positif (infeksi Shigella flexneri tanpa terapi), kontrol obat (ciprofloxacin), serta tiga kelompok perlakuan yang mendapatkan ekstrak kayu manis dalam tiga dosis berbeda. Infeksi Shigella flexneri diberikan secara intraperitoneal, kemudian dilakukan pemberian ekstrak kayu manis sesuai dosis. Apoptosis dinilai melalui ekspresi caspase-3 pada sel epitel ileocaecal junction dengan imunohistokimia. Data dianalisis menggunakan uji One Way ANOVA yang dilanjutkan dengan uji Tukey pada taraf signifikansi p < 0,05. Hasil penelitian menunjukkan adanya perbedaan bermakna ekspresi caspase-3 antar kelompok (p < 0,001), di mana pemberian ekstrak kayu manis pada dosis 2 mg, 4 mg, dan 8 mg secara signifikan menurunkan ekspresi caspase-3 dibandingkan kelompok kontrol positif, dengan tingkat penurunan yang semakin besar seiring peningkatan dosis. Di antara ketiga dosis tersebut, dosis 8 mg menunjukkan efek penekanan apoptosis paling optimal. Dengan demikian, ekstrak kayu manis berpengaruh nyata dalam menurunkan apoptosis sel epitel ileocaecal junction mencit yang diinduksi Shigella flexneri.

Shigellosis caused by Shigella flexneri is a global health problem that is associated with intestinal mucosal damage through increased epithelial cell apoptosis. Cinnamon (Cinnamomum verum) extract contains antibacterial, anti-inflammatory, and antioxidant compounds that may suppress excessive epithelial apoptosis in the gut. This study aimed to determine the effect of cinnamon extract on epithelial cell apoptosis in the ileocaecal junction of mice (Mus musculus L.) induced by Shigella flexneri. This experimental study used a post-test only control group design with a Completely Randomized Design. Female Balb/c mice were divided into a healthy control group, a negative control group (Shigella flexneri infection without therapy), a positive control group (ciprofloxacin), and three treatment groups that received different doses of cinnamon extract. Shigella flexneri was administered intraperitoneally, followed by cinnamon extract according to each dose. Apoptosis was evaluated by assessing caspase-3 expression in epithelial cells of the ileocaecal junction using immunohistochemistry. Data were analyzed using one-way ANOVA followed by Tukey’s post hoc test with a significance level of p < 0.05. The results demonstrated significant differences in caspase-3 expression among groups (p < 0.001), with cinnamon extract at doses of 2 mg, 4 mg, and 8 mg significantly reducing caspase-3 expression compared with the infected control group, showing a greater reduction with increasing doses. Among the tested doses, the 8 mg dose exhibited the most optimal effect in suppressing epithelial apoptosis. In conclusion, cinnamon extract effectively reduces apoptosis of ileocaecal junction epithelial cells in mice induced by Shigella flexneri.
