Abstract

INDONESIA:

Bacillus subtilis merupakan bakteri yang dapat menimbulkan infeksi terutama pada individu imunokompromais, sedangkan Dracaena angustifolia (daun suji) diketahui mengandung berbagai senyawa bioaktif yang secara teoretis memiliki aktivitas antibakteri. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak etanol 96% D. angustifolia terhadap B. subtilis melalui uji Konsentrasi Hambat Minimum (KHM) dan Kadar Bunuh Minimum (KBM) dengan metode mikrodilusi pada konsentrasi 1%, 2%, 4%, dan 8%. Hasil pengukuran KHM berdasarkan penurunan nilai Optical Density (OD) menunjukkan bahwa konsentrasi 1%, 2%, 4%, dan 8% masing-masing menghasilkan persentase penurunan sebesar 0%, 1,39%, 28,50%, dan 33,04%, namun seluruhnya tidak memenuhi kriteria KHM karena tidak mencapai penurunan ≥95% sesuai pedoman Schrader, Botella, dan Vaubourgeix tahun 2023. Pada uji KBM, jumlah koloni setelah perlakuan 1%, 2%, 4%, dan 8% masing-masing adalah 244×10⁻⁷ CFU/mL, 280×10⁻⁷ CFU/mL, 195,6×10⁻⁷ CFU/mL, dan 41×10⁻⁷ CFU/mL dengan persentase penurunan koloni dibanding kontrol sebesar 33,7%, 23,9%, 46,9%, dan 88,7%, namun belum memenuhi kriteria KBM yang mensyaratkan penurunan ≥99,9%, sehingga dapat disimpulkan bahwa ekstrak etanol D. angustifolia tidak menunjukkan kemampuan menghambat maupun membunuh B. subtilis pada konsentrasi 1%, 2%, 4%, dan 8% dalam kondisi penelitian ini.

ENGLISH:

Bacillus subtilis is a bacterium capable of causing infections, particularly in immunocompromised individuals. Dracaena angustifolia contains several bioactive compounds theoretically associated with antibacterial activity. This study aimed to evaluate the antibacterial effect of the 96% ethanol extract of D. angustifolia against B. subtilis using Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) assays through the microdilution method at concentrations of 1%, 2%, 4%, and 8%. MIC measurements based on Optical Density (OD) revealed inhibition percentages of 0%, 1.39%, 28.50%, and 33.04% for the 1%, 2%, 4%, and 8% concentrations, respectively. None of these values fulfilled the MIC criterion of ≥95%. In the MBC assay, colony counts following exposure to 1%, 2%, 4%, and 8% were 244×10⁻⁷, 280×10⁻⁷, 195.6×10⁻⁷, and 41×10⁻⁷, corresponding to reduction percentages of 33.7%, 23.9%, 46.9%, and 88.7%. These values did not satisfy the ≥99.9% reduction required for bactericidal activity. Therefore, the 96% ethanol extract of D. angustifolia did not exhibit inhibitory or bactericidal activity against B. subtilis at concentrations of 1%, 2%, 4%, and 8% under the conditions of this study.
