Abstract
INDONESIA:
Salmonellosis merupakan infeksi saluran pencernaan yang ditularkan melalui makanan dan masih menjadi permasalahan kesehatan baik secara global maupun nasional. Penyakit ini umumnya disebabkan oleh Salmonella enterica, khususnya serotipe Salmonella typhimurium, yang dapat menimbulkan gastroenteritis hingga komplikasi serius. Penanganan infeksi S. typhimurium semakin sulit karena kemampuan bakteri membentuk biofilm melalui komponen curli, selulosa, BapA, O-antigen, dan eDNA, yang berperan dalam meningkatkan ketahanan terhadap kondisi ekstrem serta resistensi terhadap antibiotik. Kondisi tersebut diperburuk oleh meningkatnya kasus resistensi antibiotik pada S. typhimurium, termasuk di Indonesia, sehingga diperlukan alternatif terapi yang efektif dan aman. Salah satu kandidat yang berpotensi adalah jeruk nipis (Citrus aurantifolia), khususnya bagian kulitnya yang mengandung senyawa bioaktif seperti flavonoid, tanin, saponin, alkaloid, dan minyak atsiri yang diduga berperan dalam mekanisme antibiofilm. Penelitian ini menggunakan rancangan true experimental dengan tujuh kelompok, terdiri atas tiga kelompok kontrol dan empat kelompok perlakuan dekok kulit jeruk nipis pada konsentrasi 50%, 25%, 12,5%, dan 6,25%, yang diuji menggunakan metode microtiter biofilm assay. Hasil penelitian menunjukkan bahwa dekok kulit jeruk nipis mampu mencegah perlekatan awal, menghambat pertumbuhan, serta menghancurkan biofilm matang S. typhimurium. Meskipun hasil uji pencegahan perlekatan pada konsentrasi 6,25% menunjukkan hasil yang tidak sebagaimana mestinya, temuan ini menunjukkan potensi kulit jeruk nipis sebagai agen antibiofilm alami. Oleh karena itu, penelitian lanjutan diperlukan untuk mengidentifikasi senyawa aktif spesifik serta mengoptimalkan metode ekstraksi guna mendukung pengembangan terapi alternatif antibiofilm berbasis tanaman herbal.

Salmonellosis is a gastrointestinal infection transmitted through food and remains a significant health problem both globally and nationally. This disease is generally caused by Salmonella enterica, particularly the Salmonella typhimurium serotype, which can lead to gastroenteritis and severe complications. The management of S. typhimurium infection has become increasingly difficult due to its ability to form biofilms through components such as curli, cellulose, BapA, O-antigen, and eDNA, which enhance bacterial resistance to extreme conditions and antibiotics. This condition is further aggravated by the increasing incidence of antibiotic resistance in S. typhimurium, including in Indonesia, thereby requiring an effective and safe alternative therapy. One potential candidate is lime (Citrus aurantifolia), particularly its peel, which contains bioactive compounds such as flavonoids, tannins, saponins, alkaloids, and essential oils that are presumed to play a role in antibiofilm mechanisms. This study employed a true experimental design with seven groups, consisting of three control groups and four treatment groups of lime peel decoction at concentrations of 50%, 25%, 12.5%, and 6.25%, tested using the microtiter biofilm assay method. The results demonstrated that lime peel decoction was able to prevent initial adhesion, inhibit growth, and disrupt mature biofilms of S. typhimurium. Although the adhesion prevention test at a concentration of 6.25% showed inconsistent results, these findings indicate the potential of lime peel as a natural antibiofilm agent. Therefore, further research is needed to identify specific active compounds and optimize extraction methods to support the development of herbal plant-based antibiofilm therapies.
