Abstract

ABSTRAK:

Diabetes melitus merupakan penyakit metabolik kronis yang ditandai dengan resistensi insulin dan kadar gula darah tinggi yang dapat memicu komplikasi serius termasuk gangguan fungsi hepar. Gangguan ini sering dikaitkan dengan penumpukan lemak di hepar dan stres oksidatif yang dapat merusak struktur hepar. Penelitian ini bertujuan untuk menguji pengaruh kombinasi nanopartikel
ekstrak daun bungur (Lagerstroemia speciosa), daun juwet (Syzygium cumini) dan rimpang temulawak (Curcuma xanthorrhiza) terhadap berat hepar dan hepatosomatik indeks pada hepar mencit diabetes melitus. Penelitian ini dilakukan secara eksperimental menggunakan Rancangan Acak Lengkap (RAL). Model mencit diabetes dibuat dengan pemberian diet tinggi lemak (HFD) + dextrose selama 14 hari, diikuti injeksi Streptozotocin secara intraperitoneal. Dalam penelitian ini menggunakan 24 ekor mencit BALB-C dengan 6 perlakuan dan 4 ulangan meliputi : K-N, KDM, KMET (DM + metformin), 200N (DM + nanopartikel ekstrak dosis 200mg/kgBB), 300N (DM + dosis nanopartikel ekstrak
300mg/kgBB), 300M (DM + ekstrak mikron dosis 300mg/kgBB). Analisis data
dengan uji statistik, persentase berat hepar dan hasil data HSI menggunakan analisis SPSS One way ANOVA (p<0,05). Hasil dari penelitian berat hepar didapatkan nilai signifikansi >0,05 yang artinya tidak ada perbedaan pengaruh yang signifikan terhadap beberapa kelompok perlakuan. Hasil dari penelitian nilai HSI dosis herbal yakni dosis 200mg/KgBB berpengaruh terhadap persentase
penurunan steatosis hepar dan nilai HSI.

ABSTRACT:

Diabetes mellitus is a chronic metabolic disease characterized by insulin resistance and high blood sugar levels that can trigger serious complications including impaired liver function. This disorder is often associated with fat accumulation in the liver and oxidative stress that can damage the liver structure. This study aims to test the effect of a combination of nanoparticles of bungur leaf extract (Lagerstroemia speciosa), juwet leaf (Syzygium cumini) and temulawak rhizome (Curcuma xanthorrhiza) on liver weight and hepatosomatic index in the liver of diabetic mice. This study was conducted experimentally using a Completely Randomized Design (CRD). The diabetic mouse model was made by giving a high-fat diet (HFD) + dextrose for 14 days, followed by intraperitoneal
injection of Streptozotocin. This study used 24 BALB-C mice with 6 treatments and 4 replications including: K-N, KDM, KMET (DM + metformin), 200N (DM + nanoparticle extract dose 200mg / kgBW), 300N (DM + nanoparticle extract dose 300mg / kgBW), 300M (DM + micronized extract dose 300mg / kgBW). Data analysis using statistical tests, liver weight percentage and HSI data results
using SPSS One way ANOVA analysis (p<0.05). The results of the liver weight study obtained a significance value of >0.05, which means there is no significant difference in the effect on several treatment groups. The results of the study of the HSI value of herbal doses, namely a dose of 200 mg/kgBW, have an effect on the percentage of decreased liver steatosis and HSI values.
