Abstract
ABSTRAK:
Herbal oil ekstrak daun kelor memiliki manfaat diantaranya sebagai antioksidan, antikanker, dan antimikroba karena tingginya kandungan senyawa flavonoid dan alkaloid di dalamnya. Ekstrak daun kelor dalam EVOO bersifat stabil, serta aman untuk digunakan. Tujuan penelitian ini untuk mengetahui pengaruh variasi lama ekstraksi dan volume tween 80: PEG 400 penambahan terhadap kadar total flavonoid dan alkaloid pada herbal oil ekstrak daun kelor dalam EVOO.
Tahap ekstraksi dilakukan dengan variasi waktu ekstraksi 0 menit, 20 menit pada suhu 30°C, 30 menit pada suhu 36°C, dan 40 menit pada suhu 52°C serta variasi volume tween 80: PEG 400 sebanyak 0,675 mL; 0,75 mL; 0,825 mL; dan 0,9 mL. Larutan sampel disonikasi dengan frekuensi 42 kHz. Filtrat yang telah dipisahkan menggunakan cheesecloth ditampung dan diuji kadar total flavonoid (TFC) dengan menggunakan larutan standar kuersetin, dan uji kadar total alkaloid (TAC) dengan larutan standar kafein. Selanjutnya diukur absobansi masing-masing sampel menggunakan Spektrofotometer UV-Vis untuk mengetahui kandungan total flavonoid dan alkaloid.
Hasil penelitian menunjukkan bahwa herbal oil ekstrak daun kelor dalam EVOO menghasilkan kadar flavonoid dan alkaloid tertinggi pada waktu ekstraksi 30 menit dan penambahan tween 80: PEG 400 sebanyak 0,825 mL. Kadar total flavonoid (TFC) diperoleh sebesar 103,225±0,480 mg QE/g dan kadar total alkaloid (TAC) sebesar 1,284±0,076 mg/g.
ABSTRACT:
Moringa leaf extract herbal oil has various benefits, including antioxidant, anticancer, andantimicrobial properties due to its high flavonoid and alkaloid content. The extract in EVOO is stable andsafe for use. This study aimed to determine the effect of extraction time variation and the volume ofTween 80: PEG 400 on the total flavonoid and alkaloid content in Moringa leaf extract herbal oil inEVOO.The extraction process was carried out with time variations of 0 minutes, 20 minutes at 30°C,30 minutes at 36°C, and 40 minutes at 52°C, and with varying volumes of Tween 80: PEG 400 at 0.675mL, 0.75 mL, 0.825 mL, and 0.9 mL. The sample solution was sonicated at a frequency of 42 kHz. Thefiltrate separated using cheesecloth was collected and tested for total flavonoid content (TFC) usingquercetin as the standard solution, and total alkaloid content (TAC) using caffeine as the standard. Theabsorbance of each sample was then measured using a UV-Vis spectrophotometer to determine thetotal flavonoid and alkaloid content.The results showed that the Moringa leaf extract herbal oil in EVOO had the highest flavonoidand alkaloid levels with a 30-minute extraction time and the addition of 0.825 mL of Tween 80: PEG400. The total flavonoid content (TFC) was 103.225±0.480 mg QE/g, and the total alkaloid content (TAC)was 1.284±0.076 mg/g
