Uji aktivitas ekstrak etanol 70% daun bidara Arab (Ziziphus spina-cristi) terhadap histopatologi pankreas pada tikus putih Wistar (Rattus norvegicus) yang diinduksi diet tinggi lemak

Hiperlipidemia adalah kondisi yang terjadi ketika kadar lemak (lipid) dalam darah mengalami kenaikan. Pankreas adalah salah satu organ yang terdampak kenaikan kolestrol dalam tubuh. Kenaikan kolestrol yang disebabkan oleh pola makan tinggi kolesterol menyebabkan berbagai masalah pada pankreas seperti pankreatitis akut atau diabetes. Salah satu tanaman yang memiliki banyak manfaat kesehatan adalah daun bidara (Ziziphus spina-christi), Bidara dikenal memiliki efek antioksidan dan antiinflamasi yang signifikan. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian esktrak etanol 70% daun bidara (Ziziphus spina-christi) terhadap diameter pulau langerhans dan jumlah sel acinar tikus putih wistar (Rattus norvegicus) yang diberi diet tinggi lemak. Jenis penelitian ini adalah true experimental laboratory posttest only controlled group dengan jumlah sampel hewan coba yang digunakan sebanyak 25 ekor dan dikelompokkan dalam 5 kelompok. Kelompok K- pemberian aquadest, kelompok K+ pemberian simvastatin, kelompok P1, P2, P3 pemberian ekstrak dengan variasi dosis 50,100,200 mg/kg BB tikus. Perlakuan setiap kelompok dilakukan selama 42 hari dengan 28 hari induksi lemak dan 14 hari pemberian ekstrak. Hasil uji stastistik menunjukkan adanya pengaruh ekstrak etanol 70% daun bidara dengan dosis 50 mg/kg/hari terhadap jumlah sel acinar dan dosis 200mg/kgBB tikus/hari terhadap diameter pulau langerhans. Dapat disimpulkan bahwa ektrak etanol 70% daun bidara (Ziziphus spina-christi) memliki pengaruh terhadap histopatologi pankreas tikus yang diinduksi diet tinggi lemak

Hyperlipidemia is a condition that occurs when blood lipid levels rise. The pancreas is one of the organs affected by the rise in cholesterol in the body. The increase in cholesterol caused by a high-cholesterol diet causes various problems with the pancreas such as acute pancreatitis or diabetes. One plant that has many health benefits is bidara leaf (Ziziphus spina-christi), Bidara is known to have significant antioxidant and anti-inflammatory effects. This study aims to determine the effect of 70% ethanol extract of bidara leaves (Ziziphus spina-christi) on the diameter of langerhans islets and the number of acinar cells in wistar white rats (Rattus norvegicus) fed a high-fat diet. This study is a true experimental laboratory design posttest only controlled group, with the number of experimental animal samples used as many as 25 rats and grouped into 5 groups. Group K- with the administration of aquadest, group K+ with the administration of simvastatin, group P1, P2, P3 with the administration of extracts with varying doses of 50, 100, 200 mg/kg BW rats. Treatment of each group was carried out for 42 days with 28 days of fat induction and 14 days of extract dosage. Histomorphometric observations with Hematoxylin Eosin staining to increase the number of acinar cells and diameter of the islet of langerhans using a 400x. The results showed that 70% ethanol extract of bidara leaves at a dose of 200mg/kgBB rats/day was the optimal dose to increase the number of acinar cells and the diameter of the islets of langerhans. It can be concluded that 70% ethanol extract of bidara leaves (Ziziphus spina-christi) has an effect on the histopathology of the pancreas of rats induced by a high-fat diet.

ارتفاع نسبة الدهون في الدم هو حالة تحدث عندما يرتفع مستوى الدهون (اللبيدات) في الدم. يعد البنكرياس أحد الأعضاء التي تتأثر بارتفاع نسبة الكوليسترول في الجسم. يؤدي ارتفاع نسبة الكوليسترول الناتج عن اتباع نظام غذائي يحتوي على نسبة عالية من الكوليسترول إلى حدوث مشاكل مختلفة في البنكرياس مثل التهاب البنكرياس الحاد أو مرض السكري. من النباتات التي تتمتع بفوائد صحية عديدة أوراق نبات الزيزفون (Ziziphus spina-christi). ومن المعروف أن أوراق نبات الزيزفون لها تأثيرات مضادة للأكسدة ومضادة للالتهابات. تهدف هذه الدراسة إلى تحديد تأثير إعطاء مستخلص إيثانول بنسبة %70 من أوراق نبات الزيزفون (Ziziphus spina-christi) على قطر جزر لانجرهانز وعدد الخلايا الأسينية في فئران ويستار البيضاء (Rattus norvegicus) التي تتغذى على نظام غذائي عالي الدهون. تعتبر هذه الدراسة تصميمًا تجريبيًا حقيقيًا مختبريًا لمجموعة محكومة فقط بعد الاختبار، حيث بلغ عدد عينات الحيوانات التجريبية المستخدمة ما يصل إلى 25 حيوانًا تم تجميعها في 5 مجموعات. تم إعطاء المجموعة K- الماء المقطر، وتم إعطاء المجموعة K+ سيمفاستاتين، وتم إعطاء المجموعات P1، P2، P3 مستخلصًا مع جرعات متفاوتة 50، 100، 200 ملغ/كغ من وزن الجسم للفئران. تم إجراء علاج كل مجموعة لمدة 42 يومًا بواقع 28 يومًا من حقن الدهون و14 يومًا من جرعة المستخلص. الملاحظة الهيستومورفومترية باستخدام صبغة الهيماتوكسيلين إيوزين لزيادة عدد الخلايا الأسينية باستخدام المجهر الضوئي بتكبير x400 وقطر جزر لانجرهانز باستخدام المجهر الضوئي بتكبير x400. وأظهرت نتائج الدراسة أن مستخلص الإيثانول 70% من أوراق البيدارا بجرعة 200 ملغم/كجم من وزن الجسم للفئران/يوم كانت الجرعة المثلى لزيادة عدد الخلايا الأسينية وقطر جزر لانجرهانز. يمكن الاستنتاج أن مستخلص الإيثانول بنسبة %70 من أوراق السد (Ziziphus spina-christi) له تأثير على الأنسجة المرضية في البنكرياس لدى الفئران الناتجة عن اتباع نظام غذائي عالي الدهون.
