Abstract
INDONESIA:
Hingga saat ini belum ada terapi spesifik untuk mencegah ataupun mengobati NAFLD, intervensi statin sebagai terapi NAFLD belum dapat memberikan hasil yang optimal dan dapat menimbulkan efek samping berupa hepatotoksisitas dan miopati. Oleh karena itu, penelitian ini berfokus pada pengembangan salah satu produk nutraceutical yaitu Ekstrak Fermentasi Sambal Lalapan (EFSL) sebagai kandidat terapi pengobatan NAFLD dengan memberi efek antiinflamasi pada kelinci yang diinduksi diet tinggi lemak. Tujuan Penelitian: Mengetahui pengaruh EFSL terhadap ekspresi gen relatif mRNA Interleukin-6 (IL-6) hepar pada kelinci model NAFLD. Metode: Kelinci New Zealand White jantan yang berjumlah 24 ekor dibagi menjadi 4 kelompok, yaitu kelompok dengan diet pakan standar, diet tinggi lemak, diet tinggi lemak dengan EFSL 100 mg/KgBB, dan diet tinggi lemak dengan EFSL 200 mg/Kg/BB. Ekspresi gen relatif mRNA IL-6 dikuantifikasi dengan metode PCR dan dihitung menggunakan metode Livak 2-ΔΔCT. Hasil: Dosis EFSL 100 mg/Kg/BB dapat menurunkan ekspresi gen relatif mRNA IL-6 menjadi 0,53±0,08 (p=0,000 dibandingkan dengan kelompok diet tinggi lemak) dan dosis EFSL 200 mg/Kg/BB menjadi 0,39±0,49 (p=0,000 dibandingkan dengan kelompok diet tinggi lemak). Kesimpulan: EFSL dapat menurunkan ekspresi gen relatif mRNA IL-6 oleh efek antiinflamasi yang terkandung pada senyawa fermentasi dengan target sasarannya yaitu blokade jalur inflamasi IL-6 hingga mendekati kelompok diet pakan standar.

At this moment, there is no specific therapy to prevent or treat NAFLD, statin intervention as a therapy for NAFLD has not been able to provide optimal results and can cause side effects, such as hepatotoxicity and myopathy. Therefore, this study is focusing on the development of one of the nutraceutical products, namely Fermented Vegetable Spices Extract (FVSE) as a candidate for NAFLD therapy by providing anti-inflammatory effects on rabbits induced by high-fat diet. Objective: This study is aiming to determine the effect of EFSL on the relative gene expression of hepatic Interleukin-6 (IL-6) mRNA in NAFLD model rabbits. Method: Twenty-four male New Zealand White rabbits were divided into 4 groups, groups with standard diet, high-fat diet, high-fat diet with EFSL 100 mg/KgBW, and high-fat diet with EFSL 200 mg/Kg/BW. The relative IL-6 mRNA gene expression was quantified by PCR method and calculated using the Livak 2-ΔΔCT method. Results: The 100 mg/Kg/BW EFSL dose can reduce the relative gene expression of IL-6 mRNA to 0.53±0.08 (p=0.000 compared to the high-fat diet group) and the 200 mg/Kg/BW EFSL dose can reduce to 0.39±0.49 (p=0.000 compared to the high-fat diet group). Conclusion: EFSL can reduce the relative gene expression of IL-6 mRNA by the antiinflammatory effect contained in fermented compounds that targetting the blockade of the IL-6 inflammatory pathways to approach the standard diet group.
