Pengaruh terapi buah pare (momordica charantia l) terhadap cd4 dan cd8 pada mencit balb/c yang diinfeksi plasmodium berghei

Infeksi Plasmodium dapat menyebabkan peningkatan CD4 dan CD8 abnormal akibat respon pertahanan diri sistem imun yang berlebihan dalam melawan parasit. Sistem imun dapat menjadi “pedang bermata dua” bagi sel host yang dapat mengarahkan pada kerusakan organ pada malaria berat. Buah pare (Momordica charantia L.) memiliki senyawa flavonoid, terpenoid, dan alkaloid yang bekerja sebagai anti-inflamasi dan imunoregulator dengan cara menghambat sitokin proinflamasi Th1 dan meningkatkan aktivitas IL-4 sehingga dapat menurunkan kadar CD4 dan CD8 yang berlebihan. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak buah M. charantia terhadap kadar CD4 dan CD8 mencit Balb/c yang diinfeksi Plasmodium berghei. 24 ekor mencit jantan dibagi dalam 4 kelompok sebagai berikut: Kelompok kontrol negatif (-) diinfeksi tanpa diberi terapi, kelompok perlakuan diinfeksi dan diberi terapi ekstrak buah M. charantia dengan dosis 0,25; 0,50; 1 mg/gBB/hari. Kelompok perlakuan diberi terapi selama 5 hari. Pada hari ke-8, mencit dibedah untuk diambil sampel hepar, kemudian dilakukan pengukuran kadar CD4 dan CD8 menggunakan metode imunohistokimia. Kadar CD4 dan CD8 dianalisa menggunakan One-Way ANOVA. Hasil penelitian menunjukkan bahwa terdapat perbedaan yang signifikan pada pemberian ekstrak buah pare terhadap ekspresi CD4 dan CD8 dengan nilai signifikansi 0,000 (p<0,05). Dapat disimpulkan bahwa ekstrak buah pare (Momordica charantia L.) dapat menurunkan CD4 dan CD8 pada mencit Balb/c yang Diinfeksi Plasmodium berghei. Dosis 1 mg/gBB dalam penelitian ini merupakan dosis yang optimal untuk menurunkan ekspresi CD4 dan CD8 pada mencit Balb/c yang diinfeksi Plasmdium berghei.

Plasmodium infection can cause abnormal increases on CD4 and CD8 due to the immune system's excessive self-defense response against parasites. The immune system can be a “double-edged sword” for host cells that can lead to organ damage in severe malaria. Bitter melon (Momordica charantia L.) has flavonoid, terpenoid, and alkaloid compounds that work as anti-inflammatory and immunoregulatory agents by inhibiting Th1 proinflammatory cytokines and increasing IL-4 activity so that it can reduce excessive CD4 and CD8 levels. This study aimed to determine the effect of M. charantia fruit extract on CD4 and CD8 levels of Balb/c mice infected with Plasmodium berghei. 24 male mice were divided into 4 groups as follows: The negative control group (-) was infected without treatment, the treatment group was infected and treated with M. charantia fruit extract at a dose of 0.25; 0.50; 1 mg/gBW/day. The treatment group was given therapy for 5 days. On the 8th day, the mice were dissected for liver samples, then the CD4 and CD8 levels were measured using the immunohistochemical method. CD4 and CD8 levels were analyzed using One-Way ANOVA. The results showed that there was a significant difference in the administration of bitter melon extract on the expression of CD4 and CD8 with a significance value of 0.000 (p<0.05). It can be concluded that bitter melon extract (Momordica charantia L.) can reduce CD4 and CD8 in Balb/c mice infected with Plasmodium berghei. The dose of 1 mg/gBW in this study was the optimal dose to reduce CD4 and CD8 expression in Balb/c mice infected with Plasmdium berghei.
