Abstract
INDONESIA:
Periodontitis adalah salah satu gangguan rongga mulut terbanyak di Indonesia. Periodontitis dapat berdampak serius pada kesehatan gigi serta kualitas hidup individu. Penyebab utama periodontitis adalah bakteri Porphyromonas gingivalis (P. gingivalis). Bakteri P. gingivalis memiliki faktor virulensi berupa lipopolisakarida (LPS) untuk menginfeksi jaringan periodontal. Terdeteksinya LPS P. gingivalis oleh sel imun menstimulasi respon inflamasi. Salah satu mediator penting pada saat inflamasi adalah sitokin Interleukin 6 (IL-6). Sitokin IL-6 berperan dalam inflamasi fase akut maupun fase lanjutan. Meskipun demikian, studi in vivo terkait pengaruh induksi LPS P. gingivalis terhadap ekspresi IL-6 masih terbatas. Penelitian ini bertujuan untuk membuktikan adanya pengaruh lama induksi LPS P. gingivalis terhadap ekspresi IL-6 pada jaringan periodontal Rattus norvegicus. Sampel penelitian terdiri dari 4 kelompok, yaitu K1 (kontrol negatif / tanpa induksi), K2 (induksi 7 hari), K3 (induksi 14 hari), dan K4 (induksi 21 hari). Tikus diberi perlakuan dengan menginjeksikan 10 µl larutan yang mengandung 1 mg/ml LPS P.gingivalis setiap 48 jam. Data dianalisis menggunakan uji Kruskal-Wallis, diikuti dengan uji Mann-Whitney sebagai uji lanjutan. Hasil penelitian menunjukkan bahwa terdapat perbedaan signifikan antar kelompok (p < 0,05). Uji lanjutan Mann-Whitney menunjukkan bahwa perbedaan signifikan ditemukan antara K1 dan K3 (p = 0,007), K1 dan K4 (p = 0,000), serta K2 dan K4 (p = 0,007), sedangkan perbedaan antara K1 dan K2 (p = 0,086), K2 dan K3 (p = 0,326), serta K3 dan K4 (p = 0,086) tidak signifikan. Uji korelasi Spearman menunjukkan bahwa terdapat korelasi positif antara lama induksi LPS dan ekspresi IL-6 (r = 0,939; p < 0,01). Hasil penelitian ini menunjukkan bahwa lama induksi LPS Porphyromonas gingivalis berpengaruh signifikan terhadap ekspresi IL-6 pada jaringan periodontal Rattus norvegicus.

Periodontitis is one of the most common oral disorders in Indonesia and can have serious impacts on dental health and the quality of life of individuals. The primary cause of periodontitis is the bacterium Porphyromonas gingivalis (P. gingivalis). This bacterium possesses a virulence factor in the form of lipopolysaccharide (LPS) that enables it to infect periodontal tissues. The detection of LPS from P. gingivalis by immune cells stimulates an inflammatory response. One important mediator involved in this inflammation is the cytokine Interleukin-6 (IL-6), which plays a role in both the acute and prolonged phases of inflammation. However, studies investigating the in vivo effects of LPS P. gingivalis induction on IL-6 expression remain limited. This study aims to demonstrate the effect of the duration of P. gingivalis LPS induction on IL-6 expression in the periodontal tissue of Rattus norvegicus. The study sample consisted of 4 groups: K1 (negative control/no induction), K2 (7 days induction), K3 (14 days induction), and K4 (21 days induction). Rats were treated by injecting 10 µl of a 1 mg/ml LPS P. gingivalis solution every 48 hours. Data were analyzed using the Kruskal-Wallis test, followed by Mann-Whitney post-hoc testing. The results showed significant differences between groups (p < 0.05). Mann- Whitney post-hoc testing revealed significant differences between K1 and K3 (p = 0.007), K1 and K4 (p = 0.000), and K2 and K4 (p = 0.007), while no significant differences were found between K1 and K2 (p = 0.086), K2 and K3 (p = 0.326), and K3 and K4 (p = 0.086). Spearman's correlation test showed a positive correlation between the duration of LPS induction and IL-6 expression (r = 0.939; p < 0.01) Based on these results, it can be concluded that there is a significant influence of the duration of LPS P. gingivalis induction on IL-6 expression.
