Abstract
ABSTRACT:
Kebiul seed kernel (Caesalpinia bonduc (L.) Roxb) contains steroid, saponin, and flavonoid compounds, one of which functions as an antioxidant. This study aims to identify compounds in the seed kernel that function as antioxidants and determine the category of antioxidant activity based on the IC50 value. The test was conducted using two methods, namely DPPH (2,2-diphenyl-1-picrylhydrazyl) and FRAP (Ferric Reducing Antioxidant Power). The extraction method used on kebiul seeds is UAE (UltrasonicAssisted Extraction) extraction method with 70% ethanol solvent. The thick extract obtained will be carried out phytochemical screening and the results found that the extract of whitefly seed kernel contains steroid/triterpenoid compounds, alkaloids, saponins, tannins, and flavonoid. Antioxidant activity tests using DPPH and FRAP methods were carried out at 8 different concentrations of 5 ppm, 10 ppm, 15 ppm, 20 ppm, 25 ppm, 30 ppm, 35 ppm, and 40 ppm with vitamin C as a positive control using 5 different concentrations of 1 ppm, 2 ppm, 3 ppm, 4 ppm, and 5 ppm and carried out with 3x replication. The results showed that the IC50 value using the DPPH method in a solution of kebiul seed kernel extract was 17.93 ppm with a positive control of 4.10 ppm. Meanwhile, the IC50 value using the FRAP method obtained a result of 21.79 ppm with a positive control of 17.41 ppm. Based on the data obtained, it can be concluded that the kebiul seed kernel extract has antioxidant activity with a very strong category tested in both methods, namely the DPPH and FRAP methods.

Kernel biji kebiul (Caesalpinia bonduc (L.) Roxb) mengandung senyawa steroid,saponin, dan flavonoid, dimana senyawa flavanoid sendiri berfungsi sebagai antioksidan.Penelitian ini bertujuan untuk mengidentifikasi senyawa pada kernel biji kebiul yangberfungsi sebagai antioksidan dan menentukan kategori aktivitas antioksidan berdasarkannilai IC50. Pengujian dilakukan menggunakan dua metode: DPPH (2,2-diphenyl-1-picrylhydrazyl) dan FRAP (Ferric Reducing Antioxidant Power). Metode ekstraksi yangdigunakan pada biji kebiul adalah metode ekstraksi UAE (Ultrasonic-Assisted Extraction)dengan pelarut etanol 70%. Ekstrak kental yang diperoleh akan dilakukan skriningfitokimia dan didapati hasil ekstrak kernel biji kebiul mengandung golongan senyawasteroid/triterpenoid, alkaloid, saponin, tanin, dan flavanoid. Uji aktivitas antioksidanmenggunakan metode DPPH dan FRAP dilakukan pada 8 konsentrasi yang berbeda yaitu5 ppm, 10 ppm, 15 ppm, 20 ppm, 25 ppm, 30 ppm, 35 ppm, dan 40 ppm dengan vitaminC sebagai kontrol positif menggunakan 5 konsentrasi yang berbeda yaitu 1 ppm, 2 ppm, 3ppm, 4 ppm, dan 5 ppm serta dilakukan dengan 3x replikasi.
