Abstract

INDONESIA:

Acalypha indica L.merupakan salah satu tumbuhan obat yang banyakditemukan di Indonesia. Tujuan penelitian untuk mengetahui aktivitas antikanker payudara T47D dan identifikasi senyawa aktif pada ekstrak dan fraksi daun dan akar Acalypha indica L.. Ekstraksi senyawa aktif dilakukan dengan metode maserasi menggunakan pelarut etanol 96%. Ekstrak etanol diekstraksi cair-cair menggunakan pelarut kloroform dan n-heksana. Ekstrak etanol, fraksi kloroform serta n-heksana daun dan akar Acalypha indica L.diuji aktivitas sitotoksik menggunakan metode MTT. Nilai IC50 yang diperoleh pada daun Acalypha indica L.ekstrak etanol, fraksi kloroform serta n-heksana berturut-turut adalah 122; 338; dan 255 µg/mL. Pada akar Acalypha indica L.diperoleh IC50 ekstrak etanol, fraksi kloroform serta n-heksana berturut-turut adalah 354; 215; dan 315 µg/mL. Aktivitas sitotoksik tertinggi adalah ekstrak etanol daun Acalypha indica L.yang mengandung senyawa steroid dan alkaloid. Hasil identifikasi adanya senyawa tersebut didukung dengan instrumentasi FTIR. Hasil identifikasi dugaan alkaloid dengan FTIR menunjukkan gugus spesifik berupa N-H, C-N, C=O dan C-H, sedangkan dugaan steroid menunjukkan gugus spesifik berupa =C-H siklik, C=C, C-H dan O-H.

ENGLISH:

Acalypha indica L.is one of herbal plant that can be found in Indonesia. This research aim to determine the anticancer activity of breast T47D andidentification active compounds in extracts and fractions of Acalyphaindica L.’s leaves and roots. Extraction of the active compound was conducted by maceration method using ethanol 96%. The concentrated extract of ethanol is conducted by liquid-liquid extraction using chloroform and n-hexane. The extract of ethanol, fraction of chloroform and n-hexane the Acalypha indica L.’s leaves and roots tested cytotoxic activity using MTT method. IC50 value on Acalyphaindica L.’s leaves obtained extract of ethanol, fractions of chloroform and n-hexane were 122; 338; and 255 mg/mL.Acalyphaindica L.’s roots was obtained IC50 ethanol extract, fractions of chloroform and n-hexane were 354; 215; and 315 mg/mL.The highest cytotoxic activity was extractsethanol of Acalyphaindica L.’s leaves containing steroid compounds and alkaloids. Existence identification result of these compound were supported by the use of FTIR instrumentation. Identification result of alkaloids assumption that did by the use of FTIR instrumentation indicated that, thera are specific clusters such as N-H, C-N, C=O and C-H, whereas, the steroids assumption indicated that, there are specific clusters such as =C-H cyclic, C=C, C-H and O-H.
