Abstract

INDONESIA:

Infusa merupakan metode ekstraksi yang mendekati cara masyarakat dalam membuat obat tradisional. Infusa pekat buah pare dapat digunakan sebagai obat oral diabetes mellitus. Penelitian ini bertujuan untuk mengetahui kandungan senyawa aktif dari infusa pekat buah pare dan hubungan lama terapi pada beberapa dosis terapi terhadap kadar glukosa darah tikus putih yang telah diinduksi aloksan.

Infusa pekat dibuat dengan komposisi 30 g simplisia dalam 100 mL pelarut air. Uji fitokimia dilakukan terhadap infusa pekat buah pare meliputi alkaloid, terpenoid, steroid, flavonoid, saponin dan tanin. Penelitian ini dilakukan secara invivo untuk mengetahui penurunan kadar glukosa darah selama terapi infusa pekat buah pare. Tikus dikondisikan hiperglikemik dengan menggunakan aloksan. Penelitian ini menggunakan 21 ekor tikus putih jantan yang dibagi menjadi 7 kelompok, yaitu kontrol normal, kontrol positif, dan 5 kontrol variasi dosis. Dosis yang digunakan yaitu 1 mL; 0,80 mL; 0,60 mL; 0,45 mL; 0,30 mL /200 g BB. Terapi dilakukan selama 14 hari dengan pengukuran kadar glukosa darah sewaktu pada hari ke- 2, 4, 5, 6, 8, 10, 12, dan 14 setelah terapi.

Hasil uji fitokimia infusa pekat buah pare mengandung alkaloid, tanin, saponin dan terpenoid. Terapi infusa pekat buah pare menunjukkan adanya pengaruh lama terapi terhadap penurunan kadar glukosa darah tikus yang diinduksi aloksan. Lama terapi berbanding lurus dengan penurunan kadar glukosa darah. Hasil analisis menunjukkan bahwa dari persamaan regresi dosis 0,30 mL/200 g BB lebih cepat dalam menormalkan kadar glukosa darah tikus yang diinduksi aloksan dengan persamaan y= -17,21x + 402,7. Waktu yang dibutuhkan yaitu 14,6 hari untuk mencapai kadar glukosa darah sewaktu menjadi normal.

ENGLISH:

Infusion is an extraction method that closes for the public in making traditional medicine. Concentrated infusion of bitter melon can be used as an oral medication of diabetes mellitus. This study aimed to determine the content of active compounds of concentrated infusion of bitter melon and the relation of long time of therapy on few doses to ward blood glucose levels of white rat that had induced by alloxan.

Concentrated infusion was made with a composition of 30 g of crude drug in 100 ml of water solvent. Phytochemical test was done to concentrated infusion of bitter melon that included of alkaloid, terpenoid, steroid, flavonoid, saponin and tannin. This study was performed through in vivo that was to determine the decrease in blood glucose levels during concentrated infusion therapy of bitter melon. The rat was conditioned hyperglycemic by using alloxan. This study used 21 white male rat that were divided into 7 groups: normal control, positive control, and 5 control doses. The dose1 mL; 0,80 mL; 0,60 mL; 0,45 mL; 0,30 mL / 200 g body weight. Therapy was conducted for 14 days with a measurement of blood glucose levels during days of 2, 4, 5, 6, 8, 10, 12, and 14 after therapy.

Phytochemical test results of concentrated infusion of bitter melon contained of alkaloid, tannin, saponin and terpenoid. concentrated infusion therapy of pare showed the influence of duration of therapy against the decrease of blood glucose levels of rat that was induced by alloxan. Long therapy is proportional to the blood glucose levels decrease. The analysis showed that from the dose regression equation of 0,30 mL/200 g BB was faster in normalizing blood glucose levels of rat that was induced by alloxan with the equation of y = -17,21x + 402,7. It required the time of 14,6 days to achieve normal blood glucose levels.
