Abstract
INDONESIA:
Kanker hepar merupakan salah satu penyakit yang mematikan. Pengobatan penyakit ini memerlukan biaya yang tinggi dan memberikan efek samping bagi tubuh, sehingga pemanfaatan tanaman obat menjadi salah satu pilihaan efektif dalam upaya pengobatan kanker. Ekstrak daun sirsak (Annona muricata Linn) memiliki senyawa yang disebut acetogenin yang diduga kuat sebagai anti kanker pada penelitian-penelitian sebelumnya. Ekstrak daun sirsak ini mampu meredam proliferasi sel berlebih dengan cara menghambat produksi energi berlebih pada mitokondria. Salah satu karsinogen yang dapat menyebabkan sel kanker adalah DMBA (7,12 Dimetylbenz (α) Anthracene). Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak daun sirsak terhadap kultur primer sel hepar baby hamster yang diinduksi DMBA.
Jenis penelitian yang dilakukan adalah penelitian eksperimental menggunakan Rancangan Acak Lengkap (RAL) dengan 7 perlakuan yaitu kontrol negatif, kontrol positif, perlakuan ekstrak daun sirsak konsentrasi 10 μg/mL, 20 μg/mL, 40 μg/mL, 80 μg/mL, dan 160 μg/mL terhadap sel hepar yang telah diinduksi DMBA 0,1 μg/mL selama 48 jam. Sampel dalam penelitian ini adalah sel hepar baby hamster umur 2 hari yang dikultur dalam medium DMEM 10% FBS+DMBA 0,1 μg/mL bagi kontrol positif serta perlakuan, kemudian diinkubasi dalam inkubator CO2 5% suhu 37oC selama 48 jam. Selanjutnya sampel diberi ekstrak daun sirsak dengan konsentrasi berbeda pada perlakuan selama 24 jam. Setelah selesai pemberian ekstrak daun sirsak selama 24 jam, sel hepar baby hamster diamati konfluenitas, viabilitas dan efek sitotoksitas ekstrak daun sirsak terhadap sel.
Hasil penelitian menunjukkan bahwa ekstrak etanol daun sirsak (Annona muricata Linn) berpengaruh terhadap konfluenitas, viabilitas dan sitotoksitas sel hepar baby hamster yang diinduksi DMBA 0,1 μg/mL selama 48 jam. Hasil analisis probit menunjukkan bahwa ekstrak etanol daun sirsak memiliki nilai LC50 22,3818 μg/mL sehingga termasuk sitotoksik aktif.

Liver cancer is one of a deadly disease. The Techniques Treatment of the disease is often using high cost and give side effects for the body. So the use of medicinal plants to be one effective option in the treatment of cancer. Extract of leaves of the soursop (Annonamuricata Linn) has a compound called acetogenin strongly suspected as anti cancer in previous studies.Soursop leaves extract are able to reduce excess cell proliferation by inhibiting the production of excess energy in the mitochondria. One of carcinogen that can cause cancer cells are DMBA (7.12 Dimetylbenz (α) Anthracene). This study aims to determine the effect of soursop leaves extract to cultured primary liver cells DMBA-induced hamster baby.
Type of research is experimental research using completely randomized design with 7 treatments that negative controls, positive controls, soursop leaves extract treatments concentration of 10 μg/mL, 20 μg/mL, 40 μg/mL, 80 μg/mL, and 160 μg/mL. For primary liver cell cultures that had been induced DMBA 0.1 μg/mL for 48 hours. The samples in this study were aged hamster fetal liver cells were cultured 2 days in 10% FBS DMEM medium DMBA + 0,1 μg/mL for the positive control and treatments, then incubated in 5% CO2 incubator at 37°C for 48 hours.Further samples were soursop leaves extract treatments with different concentrations for 24 hours. After completion of soursop leaf extract for 24 hours, primary cultures fetal hamster liver cells observed confluenity, viability and cytotoxicitysoursop leaves extract to the cells.
The results showed that the leaves extract ethanol of soursop (AnnonamuricataLinn) influence on confluenity, viability and cytotoxicity liver cells DMBA-induced hamster 0.1μg/mL for 48 hours. The results showed using probit analysis, that soursop leaves extract had LC50 values 22.3818 μg/mL so that can be included active cytotoxic.
