Abstract
INDONESIA:
Anting-anting merupakan gulma yang sering dijumpai dipinggir jalan yang sering dimanfaatkan sebagai obat. Hampir seluruh bagian dari tumbuhan ini mengandung golongan senyawa alkaloid. Penelitian ini bertujuan untuk mengetahui golongan senyawa alkaloid yang ada dalam ekstrak kasar alkaloid menggunakan instrumen UPLC-MS dan uji aktivitas antikanker dari ekstrak kasar, ekstrak kasar alkaloid dan isolat alkaloid hasil pemisahan KLTP terhadap sel kanker payudara T47D.
Metode ekstraksi yang digunakan adalah ekstraksi maserasi dengan pelarut metanol dan dipartisi (fraksinasi) dengan etil asetat. Ekstrak hasil maserasi dan partisi diuji fitokimia. Ekstrak kasar alkaloid hasil partisi diidentifikasi menggunakan instrument UPLC-MS. Pemisahan golongan senyawa aktif dengan KLTA untuk mengetahui eluen terbaik. Hasil partisi yang diperoleh di pisahkan dengan menggunakan KLT Preparatif dengan pelarut terbaik hasil pemisahan KLTA yaitu kloroform:metanol (9,5:0,5). Isolat alkaloid hasil KLT Preparatif yang didapat diuji aktivitas antikanker terhadap sel kanker payudara T47D secara in-vitro dengan metode MTT.
Hasil pengukuran kadar air diperoleh 6,41 %. Randemen hasil maserasi diperoleh 12,44 %. Hasil randemen partisi diperoleh 1,136 %. Berdasarkan hasil analisis dengan instrument UPLC-MS diduga ada golongan senyawa alkaloid trigonelin, berberin, penacetin, N-metil nikotinium, coptisin, evosantin, 4-aminonaptalimid, palmatin, dauricin, lysergol dan jatrorrhizine. Hasil berat isolat alkaloid pemisahan dengan KLTP adalah 4,6 mg. Hasil uji aktivitas antikanker ekstrak kasar, ekstrak kasar alkaloid, dan isolat alkaloid berturut-turut diperoleh nilai IC50 920, 670 g/mL, 611,709 g/mL, 303,061 g/mL.
ENGLISH:
Anting-anting is weeds which often found on the edge of road and often used as medicines. Almost the whole part of this plant contains a group of alkaloid compound. The purpose of this research is to know the existence of alkaloid compound in alkaloid crude extract using UPLC instrument and the anticancer activity from crude extract, alkaloid crude extract and isolate alkaloid as isolation results of KLTP against T47D cancer cell.
Extraction method that used is maceration extraction using methanol solvents and partitioned (fractionation) by etil asetat. Extract results from maceration and partition are phytochemicaly tested. Alkaloid crude extract as partition results is identified by UPLC instrument. The Active compound group is isolated by KLTA to know the best eluent. The Partition result obtained is isolated by using preparative KLT along with best solvent from the result of KLTA isolations i.e chloroform : methanol (9,5:0,5). The alkaloid isolate as the result of preparative KLT obtained is tested the anticancer activity against T47D cancer cell with in-vitro procedures by using MTT method.
Measurement results of water content is 6,38 %. Yield as maceration results is 12,44 %. Yield results of partition is 1,136 %. According the analysis result by using UPLC instrument, there is an existency supposition of trigoneline, berberine, phenacetine, N-methil nikotinium, coptisine, evosantine, 4-aminonaptalimide, phalmatine, dauricine, lysergol and jatrorrhizine alkaloid compound. The weight of isolate alkaloid which isolated by KLTP is 4,6 mg. The anticancer activity of Crude extract, alkaloid crude extract and isolate alkaloid are continually obtained at value IC50 920, 670 g/mL, 611,709 g/mL, 303,061 g/mL.
