Abstract
INDONESIA:
Daun sirsak (Annona Muricata Linn) diketahui memiliki aktivitas antikanker secara in-vitro. Disamping itu, zeolit diketahui mampu mengemban senyawa karena strukturnya yang berpori dan memiliki ruang kosong yang bisa diisi oleh molekul lain. Penelitian ini bertujuan untuk mengatahui pengaruh ekstrak etanol daun sirsak yang diembankan pada zeolit. Selain itu, untuk mengetahui rasio impregnasi paling efektif dalam menghambat pertumbuhan sel kanker secara in vitro.
Serbuk daun sirsak diekstraksi maserasi menggunakan pelarut etanol 95%. Ekstrak etanol daun sirsak yang diembankan pada zeolit NaX dengan menggunakan metode impregnasi basah. Variasi rasio ekstrak yang diembankan pada zeolit NaX yang digunakan yaitu 1:10, 5:10, dan 10:10. Masing-masing sampel hasil impregnasi, ekstrak etanol daun sirsak, dan zeolit NaX di uji aktivitas antikanker terhadap sel T47D secara in-vitro menggunakan metode MTT. Analisis FTIR juga dilakukan pada sampel hasil pengembanan untuk mengetahui perbedaan struktur zeolit sebelum dan setelah dilakukan pengembanan.
Hasil uji aktivitas antikanker menggunakan metode MTT menunjukkan nilai IC50 sampel ekstrak etanol daun sirsak sebesar 240,165 µg/mL. Pengembanan yang dilakukan belum meberikan pengaruh yang baik, karena sampel zeolit dan sampel hasil impregnasi memiliki nilai IC50 > 1000 ppm. Hal itu menunjukkan bahwa sampel tidak memiliki aktivitas menghambat pertumbuhan sel kanker T47D. Hasil analisis menggunakan FTIR menunjukkan bahwa spektra semua sampel hasil pengembanan di dominasi oleh spektra zeolit NaX. Muncul serapan baru pada bilangan gelombang 1380 cm-1 yang menunjukkan serapan gugus C-H dari ekstrak etanol daun sirsak. Pada sampel 5:10 dan 10:10 ada spektra yang mengalami pelebaran pita dan peningkatan intensitas serapan.
ENGLISH:
The Soursop leaf (Annona muricata Linn) is known to have in vitro anticancer activity. Beside that, zeolites can carry anticancer compounds because the porous and the space of the zeolite structure that can filled with another compound. This study aims to know the influence of ethanol extract of soursop leaf that impregnated to zeolite NaX. Beside that to know which ratio are most effective in inhibiting the growth of cancer cells.
The soursop leaf powder was extracted using ethanol 95 %. The ethanol extract of soursop leaf was loaded to zeolite NaX using impregnation method. Variations of soursop leaf extract that loaded to NaX zeolite used are 1:10, 5:10, and 10:10. Each impregnated sampel, ethanol extract of soursop leaf, and zeolite NaX were tested the anticancer activity to T47D cell line in vitro using MTT method. FTIR analysis also perfomed on the impregnated sampel to determine the differences of zeolite structure before and after impregnated.
The results of the anticancer activity test using MTT method showed that the IC50 value of ethanol extract of soursop leaf is 240,165 µg / mL. The impregnation showed negative influence to the anticancer activity, because the IC50 value of the NaX zeolite and impregnated sample was higher than 1000 ppm. It shows that the samples had no activity in inhibiting the growth of T47D cancer cell. The FTIR analysis showed that the spectra of all impregnated sample was dominated by the spectra of NaX zeolite structure. A new absoption appear at 1380 cm-1 indicates thr C-H group absorption from ethanol extract of soursop leaf. The spectra of sample ratio 5:10 and 10:10 there a band broadening and increase absorption intensity.
