Abstract
INDONESIA:
Demam tifoid merupakan penyakit infeksi saluran pencernaan yang disebabkan oleh bakteri S. typhi. Daun pepaya gunung (C. pubescens) memiliki potensi sebagai antibakteri untuk pengobatan demam tifoid. Penelitian ini dilakukan untuk mengetahui potensi aktivitas antibakteri hasil fraksi etil asetat ekstrak etanolik daun pepaya gunung secara in vitro dan potensi senyawa aktif pada daun pepaya gunung dalam menghambat aktivitas DNA gyrase. Penelitian ini bersifat eksperimental. Proses ekstraksi menggunakan metode maserasi dan fraksinasi menggunakan metode ekstraksi cair-cair. Uji antibakteri fraksi etil asetat daun pepaya gunung dilakukan pada konsentrasi 10%, 25%, 50%, 75%, dan 100% dengan empat kali pengulangan. Metode pengujian menggunakan difusi sumur agar. Kontrol positif menggunakan antibiotik ciprofloxacin dan kontrol negatif menggunakan pelarut ekstrak DMSO. Analisis data diameter daerah hambat menggunakan one way ANOVA. Uji in silico dilakukan dengan menggunakan senyawa carpaine, chlorogenic acid, kaempferol, mirycetin, caffeic acid, dan ferulic acid sebagai ligan dan DNA gyrase subunit B sebagai reseptor. Uji in silico dilakukan dengan tiga tahapan yakni prediksi aktivitas antibakteri, lipinski role of five, dan penambatan molekul (molecukar docking) melalui software pyrx. Konsentrasi terbaik uji in vitro adalah 100% dengan daya hambat 18,72 mm (P<0,05). Senyawa pada daun pepaya gunung dapat bekerja aktif secara oral karena memenuhi semua aturan Ro5. Hasil docking myriceti dan kaemferol memiliki nilai energi ikatan terbaik dibandingkan dengan antibiotik ciprofloxacin, yaitu -15,4 kkal/mol. Sehingga senyawa pada daun pepaya gunung berpotensi sebagai antibiotik.
ENGLISH:
Typhoid fever is a disease of the gastrointestinal tract infections caused by S. typhi bacteria. Mountain papaya leaves (C. pubescens) has potential as an antibacterial for the treatment of typhoid fever. This research was conducted to understand the potential of antibacterial activity from ethyl acetate fraction of mountain papaya lef ethanolic extract in vitro and potency of active compounds in the mountain papaya leaf in inhibiting the activity of DNA gyrase. This research has quality is experimental. The process of extraction uses maceration and fractionation method using liquid-liquid extraction methods. Antibacterial Test ethyl acetate fraction of mountain papaya leaf conducted at a concentration of 10%, 25%, 50%, 75%, and 100% with four repetitions. The agar well diffusion method is used for antibacterial activuty test. Positive controls using the antibiotic ciprofloxacin and negative controls using the solvent DMSO extract. Inhibition area diameter data analysis using one-way ANOVA. Test in silico did used carpaine compound, chlorogenic acid, kaempferol, mirycetin, caffeic acid and ferulic acid as ligands and DNA gyrase subunit B as a receptor. In silico did this test with three phases namely antibacterial activity prediction, Lipinski role of five, and belay molecule (molecular docking) through software PyRx. The best concentration in vitro test was 100% with the inhibition of 18.72 mm (P <0.05). Compounds in mountain papaya leaves can work orally active because it full fill all the rules Ro5. Results of docking myriceti and kaempferol have the best bond energy scare compared to the antibiotic ciprofloxacin, which is -15.4 kcal/mol. That’s why the compound on the mountain papaya leaf potentially as antibiotics.
